目的:研究凋亡相关因子Caspase-3 和Bcl-2 在甲基苯丙胺神经毒性机制中所发挥的作用。方法:40 只健康雄性SD 大鼠随
机分为对照组(n=10)和实验组(又分为3个亚组,分别为注射1 天后、4 天后、7 天后,n=10)。实验组给予20mg/kg的甲基苯丙胺腹
腔注射,对照组给予相同剂量的生理盐水注射。用免疫组化检测中毒大鼠相关脑区Caspase-3 和Bcl-2 的表达,用图像分析技术对
检测结果进行分析。结果:Caspase-3 在中毒大鼠不同脑区表达逐渐增加并可见明显的阳性信号;Bcl-2 在中毒大鼠不同脑区表达
逐渐减弱。结论:凋亡相关因子Caspase-3 和Bcl-2 参与了甲基苯丙胺神经毒性机制。 |

Objective: To study the function of Caspase-3 and Bcl-2 protein in the methamphetamine mechanism. Methods: Forty
male Sprague-Dawley rats were randomly divided into control group (n=10) and experimental group(dividing it into the first, fourth and
seventh group after the injection, n=10). Rats in the experimental group were intraperitoneal injected with methamphetamine hydrochloride
(20mg/kg), and those in the control group were injected with saline with the same volume. Then examining the protein expression of
Caspase-3 and Bcl-2 of poisoning rats in related encephalic region by immunohistochemistry technique and analyzing the test results
through image. Results: Expression of Caspase-3 increased significantly in the related encephalicregion in rat poisoning of methamphetamine
where exist more clearly positive signal. Express of Bcl-2 the reduced gruadually in the related encephalicregion in rat poisoning
of methamphetamine. Conclusion: The abnormal expression of Caspase-3 and Bcl-2 shows that they take part in the Methamphetamine-
Induced Neurotoxicity. |
