本研究基于转录组学技术探讨多发性抽动症(Tourette syndrome, TS)模型大鼠的关键miRNA-mRNA调控网络及潜在中药干预。首先通过亚氨基二丙腈构建TS大鼠模型,再通过RNA测序筛选TS大鼠脑组织差异表达的miRNA与mRNA,结合生物信息学分析构建互作网络,进一步通过网络药理学筛选潜在中药。结果表明造模7天后,模型组出现运动行为及刻板改变,模型组行为学评分大于3分,HE染色可见部分神经元细胞核形态不规则,染色质分布不均匀,存在核固缩现象,胶质细胞数量增多。KEGG富集分析提示关键信号通路:钙离子信号通路、神经活性配体-受体相互作用、p53信号通路、ECM-受体相互作用、TGF-β信号通路,而miR-125a-3p、miR-106-3p、miR-760-3p可能是关键的miRNA。潜在中药:筋骨草、五加皮、党参、千金藤、龙骨、羌活、罗汉果、面针根、巴戟天、延胡索。本研究初步揭示了IDPN诱导TS模型大鼠的关键分子机制及中药干预的潜在靶点。 |

: The study explores the key miRNA mRNA regulatory network and potential traditional Chinese medicine intervention in Tourette syndrome (TS) model rats based on transcriptomics technology. Firstly, the TS rat model was constructed using iminodipropionitrile, and then differentially expressed miRNAs and mRNAs in the brain tissue of TS rats were screened by RNA sequencing. Combined with bioinformatics analysis, an interaction network was constructed, and potential traditional Chinese medicines were further screened through network pharmacology. The results showed that after 7 days of modeling, the model group exhibited changes in motor behavior and rigidity. The behavioral score of the model group was greater than 3 points, and HE staining revealed irregular nuclear morphology, uneven distribution of chromatin, nuclear condensation, and an increase in the number of glial cells in some neurons. KEGG enrichment analysis suggests key signaling pathways: calcium ion signaling pathway, neuroactive ligand receptor interaction, p53 signaling pathway, ECM receptor interaction, TGF - β signaling pathway, while miR-125a-3p, miR-106-3p, miR-760-3p may be key miRNAs. Potential Chinese medicines: Ajuga decumbens, acanthopanax bark, Codonopsis pilosula, Stephania Root, Os Draconis, Notopterygium incisum, Siraitia grosvenorii, Zanthoxylum nitidum, Morinda officinalis, Corydalis Rhizome. The study preliminarily revealed the key molecular mechanisms of IDPN induced TS model rats and potential targets for traditional Chinese medicine intervention. |
