摘要 目的:探讨不明原因复发性流产(URSA)患者绒毛组织微小核糖核酸(miR)-146a-5p、miR-146b-5p表达与血管新生因子及微血管密度(MVD)的关系。方法:选取2020年1月-2023年12月我院收治的119例URSA患者纳入URSA组,同期选取90例正常早孕并自愿终止妊娠的妇女纳入对照组。比较两组绒毛组织miR-146a-5p、miR-146b-5p、血管新生因子[缺氧诱导因子-1α(HIF-1α)、血管内皮生长因子(VEGF)]、MVD表达。Pearson检验分析绒毛组织miR-146a-5p、miR-146b-5p表达与HIF-1α、VEGF表达及MVD的相关性。结果:相比于对照组,URSA组绒毛组织miR-146a-5p表达水平降低(P<0.05),miR-146b-5p表达水平升高(P<0.05);相比于对照组,URSA组绒毛MVD降低(P<0.05);相比于对照组,URSA组绒毛组织HIF-1α、VEGF表达水平降低(P<0.05);Pearson检验显示,URSA患者绒毛组织miR-146a-5p表达与HIF-1α、VEGF、MVD呈正相关(P<0.05),miR-146b-5p表达与HIF-1α、VEGF、MVD呈负相关(P<0.05)。结论:URSA患者绒毛组织miR-146a-5p低表达、miR-146b-5p高表达与血管新生因子HIF-1α、VEGF表达下调及MVD降低有关。 |

ABSTRACT Objective: To investigate the relationship between the expression of microRNA (miR) -146a-5p and miR-146b-5p in villus tissue of patients with unexplained recurrent spontaneous abortion (URSA) and angiogenesis factors and microvessel density (MVD). Methods: 119 URSA patients who admitted to our hospital from January 2020 to December 2023 were selected as URSA group, and 90 women with normal early pregnancy who voluntarily requested abortion to terminate pregnancy were selected as control group. The expressions of miR-146a-5p, miR-146b-5p, angiogenesis factors [hypoxia-inducible factor-1α (HIF-1α), vascular endothelial growth factor (VEGF)] and MVD in villus tissues were compared between two groups. The correlation between miR-146a-5p, miR-146b-5p expression and HIF-1α, VEGF expression and MVD in villus tissue were analyzed by Pearson test. Results: Compared with control group, the expression level of miR-146a-5p was decreased in villus tissue of URSA group (P<0.05), and the expression level of miR-146b-5p was increased(P<0.05). Compared with control group, the MVD of villi in URSA group was decreased (P<0.05). The expression levels of HIF-1α and VEGF in villus tissues of URSA group were lower than those in control group (P<0.05). Pearson test showed that, the expression of miR-146a-5p in villus tissue of URSA patients was positively correlated with HIF-1α, VEGF and MVD (P<0.05), and the expression of miR-146b-5p was negatively correlated with HIF-1α, VEGF and MVD (P<0.05). Conclusion: The low expression of miR-146a-5p and the high expression of miR-146b-5p in villous tissues of URSA patients are related to the down-regulation of angiogenesis factors HIF-1α and VEGF and the decrease of MVD. |
