Objective:To investigate the relationship between serum long non-coding RNA (lncRNA) nicotinamide nucleotide antisense transferase RNA1 (NNT-AS1), microRNA (miR)-582-5p and airway inflammation and airway hyperresponsiveness in children with bronchial asthma.Methods: 120 children with bronchial asthma who were treated in our hospital from February 2021 to August 2023 were selected, and they divided into remission group (n=52) and attack group (n=68) according to the relevant guidelines.Additionally, a control group of sixty healthy kids who had physical exams performed at our hospital at that time were chosen. The relative expression of serum lncRNA NNT-AS1, miR-582-5p and serum airway inflammatory cytokines [interleukin-17 (IL-17), soluble human stromal lysin 2 (sST2), tumor necrosis factor-α (TNF-α)], airway responsiveness indicators [basic respiratory resistance (Rrs cont), minimum induced cumulative dose (Dmin), basic respiratory conductivity (Grs cont), specific airway conduction decreased by 35 th percentile (PD35)] were compared among three groups. The relationship between serum lncRNA NNT-AS1, miR-582-5p expression and airway inflammatory cytokines, airway responsiveness indicators were analyzed by Pearson correlation test. Using a receiver operating characteristic (ROC) curve, the predictive value of serum lncRNA NNT-AS1 and miR-582-5p for bronchial asthma was examined. Results: The relative expression of lncRNA NNT-AS1, serum levels of IL-17, TNF-α, sST2 and Rrs cont in remission group and attack group were higher than those in control group, and those in attack group were higher than those in remission group (P<0.05). The relative expression of miR-582-5p, Grs cont, Dmin and PD35 in remission group and attack group were lower than those in control group, and those in attack group were lower than those in remission group (P<0.05). The expression of serum lncRNA NNT-AS1 in bronchial asthma children was positively correlated with IL-7, TNF-α, sST2 and Rrs cont (r>0, P<0.05), and negatively correlated with Grs cont, Dmin and PD35 (r<0, P<0.05). The expression of serum miR-582-5p in bronchial asthma children was negatively correlated with IL-17, TNF-α, sST2 and Rrs cont (r<0, P<0.05), and positively correlated with Grs cont, Dmin and PD35 (r>0, P<0.05). The area under the curve (AUC) of serum lncRNA NNT-AS1, miR-582-5p single and combined prediction of bronchial asthma was 0.917, 0.883 and 0.959, respectively, the combined prediction was better than single prediction.Conclusion:Serum lncRNA NNT-AS1 is highly expressed and miR-582-5p is lowly expressed in children with bronchial asthma, the expression of both are closely related to asthma stage, airway inflammation and airway hyperresponsiveness, and serum lncRNA NNT-AS1 combined with miR-582-5p can assist in predicting the occurrence of bronchial asthma. |
