摘要:活化素受体样激酶 1 ( Activin receptor-like kinase 1, ALK 1)是转化生长因子β(Transforming growth factor β, TGF-β)超家族的 I 型受体。ALK 1 表达于多个物种中,而且特异的高表达于内皮细胞和血管发生旺盛的组织,在血管网络的形成和稳态维持过程中发挥着重要作用。ALK 1 基因的异常表达会导致遗传性出血性毛细血管扩张症( Hereditary hemorrhagic telangiectasia, HHT)在内的多种疾病。本文旨在对 ALK 1 信号传导通路,ALK 1 蛋白结构,ALK 1 的功能,ALK 1 异常导致的疾病以及基于 ALK 1 的抗血管生成疗法等方面进行阐述,希望可以对未来 ALK 1 结构,功能的研究以及以 ALK 1 为靶向的药物开发有所帮助。 |

ABSTRACT: Activin receptor-like kinase 1 (ALK 1) is a type I receptor in the transforming growth factor-beta (TGF-β) super family. ALK 1 is expressed in several species and is particularly highly expressed in endothelial cells and vasogenic tissues. ALK1 plays an important role in in the formation and maintenance of vascular system. Abnormal expression of ALK 1 gene can lead to Hereditary hemorrhagic telangiectasia (HHT) and other diseases.This paper aims to elaborate on ALK 1 signaling pathway, ALK 1 protein structure, ALK 1 function, ALK 1 abnormal diseases and ALK 1 based anti-angiogenesis therapy.It is hoped that it will be helpful for the study of the structure and function of ALK 1 and the development of drugs targeting ALK-1 in the future. |
