      摘要 目的:分析黄芪提取液对呼吸道合胞病毒(RSV)感染小鼠的作用机制。方法:将30只SD小鼠随机分为对照组、模型组和黄芪组各10只。模型组和黄芪组均滴鼻滴度为10-3.5 TCID50/mL的RSV病毒液,制备小鼠RSV模型,对照组滴鼻等量生理盐水。黄芪组腹腔注射黄芪注射液18 g/kg,每日1次,连用9 d;对照组和模型组小鼠腹腔注射等体积的蒸馏水。治疗后,检测小鼠血清白细胞介素(IL)-2、IL-23、IL-17、IL-6、干扰素(IFN)-γ、超氧化物歧化酶(SOD)、丙二醛(MDA)和一氧化氮(NO)水平,Western blot检测Toll样受体(TLR)7蛋白表达水平,HE染色观察肺组织形态。结果:与对照组相比,模型组小鼠IL-23、IL-17、IL-6、NO、MDA和TLR7水平显著升高(P<0.05),IL-2、SOD和IFN-γ水平显著下降(P<0.05);与模型组相比,黄芪组小鼠的IL-23、IL-17、IL-6、NO、MDA和TLR7水平显著下降(P<0.05),IL-2、SOD和IFN-γ水平显著升高(P<0.05)。结论:黄芪提取液可以有效调节辅助性T细胞释放细胞因子,保护呼吸道合胞病毒感染引起的免疫系统功能紊乱。

      ABSTRACT Objective: To analyze the mechanism of action of Astragalus membranaceus (Huangqi) extract on mice infected with respiratory syncytial virus (RSV). Methods: 30 SD mice were randomly divided into control group, model group and Huangqi group. Model group and Huangqi group were given RSV virus solution with a nasal titer of 10-3.5 TCID50/mL, and a mouse RSV model was prepared. Control group was given the same amount of normal saline. Huangqi group was intraperitoneally injected with 18 g/kg A. membranaceus injection once/d for 9 d. Control group and model group were injected intraperitoneally with an equal volume of distilled water. After treatment, the levels of serum interleukin (IL)-2, IL-23, IL-17, IL-6, interferon (IFN)-γ, superoxide dismutase (SOD), mal- ondialdehyde (MDA) and nitric oxide (NO) were measured in mice. And the expression level of Toll-like receptor (TLR) 7 protein was detected by Western blot, and the morphology of lung tissue was observed by HE staining. Results: Compared with control group, the levels of IL-23, IL-17, IL-6, NO, MDA and TLR7 in model group were significantly increased (P<0.05) while the levels of IL-2, SOD and IFN-γ were significantly decreased (P<0.05). Compared with model group, the levels of IL-23, IL-17, IL-6, NO, MDA and TLR7 in Huangqi group were significantly decreased (P<0.05) while the levels of IL-2, SOD and IFN-γ were significantly increased (P<0.05). Conclusion: A. membranaceus extract can effectively regulate the release of cytokines by helper T cells and protect the immune system dysfunction caused by respiratory syncytial virus infection.
