      摘要 目的:探讨高脂喂养时小鼠肝脏组织的HSP70 mRNA表达量及维生素C对应激反应过程中HSP70 mRNA表达的影响,探索维生素C在高脂喂养条件下对肝脏生理性应激反应的保护作用。方法:将30只雄性C57BL/6小鼠随机分为三组,A组普通喂养组、B组高脂喂养组、C组高脂喂养+维生素C,观察三组小鼠的肝脏组织形态学差异,实时定量检测小鼠肝脏HSP70mRNA的表达量。结果:小鼠体重及肝脏指数在高脂喂养组显著增高,由的4.42%上升至8.76%,而维生素C的加入可以明显削弱这一影响,肝脏指数为5.5%,同时高脂喂养小鼠肝脏的HSP70mRNA的相对表达量为正常小鼠的3倍左右,而维生素C的加入可将这一比例降至2倍,组间两两比较有统计学差异(P<0.05)。结论:维生素C可以有效降低高脂喂养小鼠肝脏HSP70mRNA表达,并可降低高脂喂养对肝脏造成的应激损伤,提示我们维生素C减缓高脂饮食对小鼠肝脏的损伤与机体本身的应激反应有关,但其作用机制还有待进一步研究。

      ABSTRACT Objective: To investigate the effect of vitamin C on the expression of HSP70 mRNA in the liver of mice fed with high-fat diet and the effect of vitamin C on the expression of Hsp70 mRNA in response to stress. The effects of vitamin C on the physiological stress response effect. Methods: Thirty male C57BL / 6 mice were randomly divided into three groups: normal feeding group (group A), high fat feeding group (group B), high fat diet plus vitamin C (group C), liver histomorphology, And the expression of Hsp70 mRNA was detected by real-time quantitative RT-PCR. Results: The body weight and liver index of mice were significantly increased in high-fat fed group, up from 4.42% to 8.76%, while the addition of vitamin C could significantly attenuate this effect. The relative expression of Hsp70mRNA in the liver of high fat fed mice was about three times that of normal mice, butthis ratio could be reduced to2 times when add Vitamin C in diet, and there was significant difference between any of the two groups (P <0.05). Conclusion: Vitamin C can effectively reduce the expression of HSP70mRNA in the liver of mice fed with high-fat diet, and reduce the stress injury caused by by high fat diet. It suggests that vitamin C can alleviate the damage of liver and the stress response,but its mechanism remains to be further studied.
