摘要 目的:CT或MR淋巴显像技术(CT/MR-LG)对前哨淋巴结(SLN)性质的评估还存在不足之处,而如果将这两种技术结合起来则有可能提高评估的准确性。在前期研究中我们已经验证纳米探针BaGdF5-PEG可以作为CT-LG造影剂。在本研究中,我们将继续探讨纳米探针BaGdF5-PEG作为MR-LG新型造影剂显像兔乳腺SLN的可行性。方法:采用水热法制备具有良好水溶性及生物相容性的纳米探针BaGdF5-PEG。采用6只健康雌性新西兰白兔作为实验对象,于每只兔右侧第2乳晕皮下注射0.1 mL BaGdF5-PEG,所有实验兔均行两次T1加权成像,分别为造影剂注射前以及注射后20 min;将从注射点至腋窝方向上的淋巴引流通路中最先增强的淋巴结定义为SLN,记录MR-LG图像显示的所有SLN,并将MR-LG结果与前哨淋巴结活检(SLNB)对照。结果:皮下注射BaGdF5-PEG后20 min对实验兔行MR-LG,所获图像显示从注射点至腋窝方向上的淋巴引流通路中均可见明显增强的SLN,每只实验兔各显像一枚SLN,且MR-LG结果与SLNB结果一致。结论:纳米探针BaGdF5-PEG作为兔乳腺SLN的MR淋巴显像剂是可行的。 |

ABSTRACT Objective: There are still some deficiencies in the evaluation of the properties of sentinel lymph node (SLN) by CT or MR lymphography(CT/MR-LG). However, if these two techniques are combined, it is possible to improve the accuracy of the evaluation of SLN. In previous studies, we have shown that BaGdF5-PEG nanoprobes could be used as contrast agents of CT-LG. In this study, we investigated the feasibility of using BaGdF5-PEG nanoprobes as the novel MR imaging agents for the detection of SLN in rabbit mammary gland. Methods: BaGdF5-PEG nanoprobe with biological compatibility and water solubility was synthesized by the hydrothermal method and further modified using HS-PEG2000-NH2 as stabilizing agents. Six healthy female New Zealand rabbits underwent T1WI imaging after subcutaneous injection of 0.1 mL of BaGdF5-PEG in 1 sub-areolar area. MR images were taken before and 20 minutes after contrast injection. On MR-LG images, the first enhanced lymph node in the lymphatic drainage pathway was defined as the SLN. Records of the SLN were identified by the MR image and the results of MR-LG were compared with the results of SLN biopsy. Results: MR-LG was performed on the experimental rabbits after subcutaneous injection of BaGdF5-PEG nanoprobes, and all images showed enhanced SLN in the lymphatic drainage pathway from the injection site to the axillary direction. MR-LG with BaGdF5-PEG nanoprobes enabled the visualization of 6 enhanced LNs (mean = 1/rabbit). The results of MR-LG were consistent with SLNB. Conclusion: BaGdF5-PEG nanoprobes could be used as a contrast agent of MR-LG. |
