      摘要 目的:研究褪黑素(MT)对急性百草枯(PQ)中毒大鼠血浆核因子-κB(NF-κB)、肿瘤坏死因子-α(TNF-α)及白细胞介素-6(IL-6)水平的影响。方法:将75只成年雄性SD大鼠按照随机数字表法分成空白对照组、中毒组、观察组,每组各25只。中毒组及观察组大鼠腹腔注射25 mg/kg的PQ构建染毒模型,同时观察组给予10 mg/kg的MT干预,中毒组给予等量的生理盐水干预。比较1、3、10、20、30 d各组大鼠血浆NF-κB、TNF-α、IL-6水平、肾组织细胞凋亡率以及尿蛋白水平,分析观察组PQ中毒大鼠血浆NF-κB、TNF-α及IL-6水平与肾组织细胞凋亡率和尿蛋白的相关性。结果:观察组和中毒组在1~30 d的血浆NF-κB、TNF-α及IL-6水平均分别高于对照组,且观察组低于中毒组,差异均有统计学意义(均P<0.05)。观察组和中毒组在1~30d的肾组织细胞凋亡率和尿蛋白水平均分别高于对照组,且观察组低于中毒组,差异均有统计学意义(均P<0.05)。Spearman相关性分析结果显示,观察组PQ中毒大鼠血浆NF-κB、TNF-α及IL-6水平与肾组织细胞凋亡率和尿蛋白水平均呈正相关(P<0.05)。结论:应用MT对急性PQ中毒大鼠进行干预后,其血浆NF-κB、TNF-α及IL-6水平明显降低,且肾组织损害也得到缓解,其肾组织损伤与其炎症因子及氧化应激状态密切相关。

      ABSTRACT Objective: To study the effect of melatonin(MT) on plasma levels of nuclear factor-κB (NF-κB), tumor necrosis factor-α (TNF-α) and interleukin -6 (IL-6) levels in rats with acute paraquat poisoning(PQ). Methods: A total of 75 adult male SD rats were randomly divided into blank control group(n=25), poisoning group(n=25) and observation group(n=25). The rats in poisoning group and observation group were given intraperitoneal injection of 25 mg/kg PQ to construct exposure model, the observation group was given 10 mg/kg intervention by MT at the same time, the poisoning group was given equal amount of physiological saline intervention. The levels of NF-κB, TNF-α, IL-6, renal cell apoptosis rate and urinary protein in the rats of each group were compared after 1 d, 3 d, 10 d, 20 d, 30 d. The correlation between the levels of NF-κB, TNF-α, IL-6 and the cell apoptosis rate and urinary protein in renal tissue of rats with PQ poisoning was analyzed. Results: The plasma levels of NF-κB, TNF-α and IL-6 in the observation group and poisoning group at 1 d to 30 d were higher than those in the control group respectively, and the above indexes in the observation group were lower than those in the poisoning group, the differences were statistically significant (all P<0.05). The cell apoptosis rate and urinary protein level in the renal tissues of the observation group and the poisoning group at 1 d to 30 d were higher than those of the control group, and the above indexes of the observation group were lower than those of the poisoning group, the differences were statistically significant (all P<0.05). Spearman correlation analysis showed that the plasma levels of NF-κB, TNF-α and IL-6 in the PQ poisoning group were positively correlated with the cell apoptosis rate and urinary protein levels in renal tissue (P<0.05). Conclusion: After MT treatment of acute PQ poisoning rats,the plasma levels of NF-κB, TNF-α and IL-6 is significantly decreased, and the damage of renal tissue is also alleviated; the damage of renal tissue is closely related to inflammatory factors and oxidative stress.
