摘要:蛋白质组学的研究是生命科学进入后基因时代的特征,是探索生命奥秘的有效手段,相关的实验技术已被广泛应用于各个领域。传统的蛋白组学技术包括凝胶电泳、质谱分析、蛋白质芯片等,但由于这些技术存在灵敏度低、操作繁琐、可重复性差等缺点已无法适应当今蛋白质研究的需要;这就促使了蛋白组学技术的改进和革新。同位素标记的相对和绝对定量(iTRAQ )技术是目前进行蛋白定性定量常用的、发展较快的新技术,该技术灵敏度高、准确率和可重复性好,能够同时检测多个样本,并且对样本进行精准高效的分析。文章梳理了iTRAQ技术的优缺点,对该技术近几年在医药基础和临床研究应用的情况进行整理和分析,探讨该技术在医药领域的应用前景。 |

ABSTRACT: The study of proteomics is the feature of life science in the post genomic era, an effective means to explore the mys- tery of life, and related experimental technologies have been widely used in various fields. Traditional proteomic technologies are that in- clude gel electrophoresis, mass spectrometry analysis, protein chip and so on. Due to these technologies existence some deficiencies, i.e. low sensitivity, complicated operation, the disadvantages of poor repeatability, they have not been adapt to the current protein research, which prompted the improvement and innovation of protein group science and technology. Isotope labeled for relative and absolute quan- titation (iTRAQ) technology is used for qualitative and quantitative protein commonly, development as a new technology quickly, the technology has the advantages of high sensitivity, accuracy and good reproducibility, which can simultaneously detect multiple samples and the samples can be analysed accurdately and efficiently. In this paper, the advantages and disadvantages of iTRAQ technology are analyzed, the medicine and clinical researches that used the iTRAQ technology in recent years are analyzed, and the application prospects of this technology in the field of medicine are discussed. |
