      摘要 目的:研究microRNA-221及E-cadherin在胃癌组织中的表达及其临床意义。方法:选取哈尔滨医科大学附属四院2014年-2016年收治的的胃癌患者50例,分别取胃癌及癌旁正常组织作为研究组及对照组。分别应用实时荧光定量PCR及免疫组织化学方法检测其中microRNA-221及E-cadherin的表达。结果:microRNA-221在胃癌组织中的表达较正常组织显著增高(P<0.05),E-cadherin在胃癌组织中的阳性表达率较正常组织显著降低。胃癌组织中microRNA-221的表达与患者的TNM分期、淋巴结转移及远处转移具有显著相关性(P<0.05),E-cadherin的表达与患者淋巴结转移及远处转移具有显著相关性(P<0.05)。胃癌患者中E-cadherin阴性表达者microRNA-221的相对表达量明显高于E-cadherin阳性表达者(P<0.05)。结论:microRNA-221在胃癌组织中呈高表达,E-cadherin在胃癌组织中呈低表达,二者在胃癌组织中的表达呈负相关。microRNA-221可能通过影响上皮细胞间充质转化影响胃癌的浸润与转移。

      ABSTRACT Objective: To investigate the expressions and clinical significances of microRNA-221 and E-cadherin in the gastric cancer. Methods: The tumor tissues and adjacent normal tissues from 50 cases of patients with gastric cancer from the fourth affiliated hospital of Harbin medical university from 2014 to 2016 were selected as the study group and the control group respectively. The expres- sions of microRNA-221 and E-cadherin in both groups were examined by real-time RT-PCR and immunohistochemistry. Results: The expression of microRNA-221 in the gastric cancer samples was significantly higher than that of the adjacent normal samples(P<0.05). The positive expression rate of E-cadherin in the gastric cancer tissues was obviously lower than that of the adjacent normal tissues. The expression of microRNA-221 in the gastric cancer was obviously associated with the TNM stage, lymph node metastasis and distant metastasis(P<0.05). E-cadherin expression was significantly associated with the lymph node metastasis and distant metastasis of gastric cancer(P<0.05). The expression of microRNA-221 in which had negative expression of E-cadherin was obviously higher than that with positive expression of E-cadherin in the gastric cancer(P<0.05). Conclusion: The expression of microRNA-221 was high and expression of E-cadherin was low in the gastric cancer which might be associated with the invasion and metastasis of gastric cancer. microRNA-221 might participate in the invasion and metastasis of gastric cancer by Epithelial-mesenchymal transition.
