      摘要 目的:探讨三黄泻心汤对胃癌细胞增殖的影响和机制。方法:以体外培养的胃癌细胞系HGC-27为研究对象,给予三黄泻心汤处理后,采用流式细胞仪检测三黄泻心汤对胃癌细胞系HGC-27细胞周期的影响,免疫荧光检测HGC-27细胞Ki67的表达,定量PCR实验检测增殖相关非编码RNA H19表达的影响。结果:当没有幽门螺旋杆菌时,三黄泻心汤对胃癌细胞系的细胞周期、Ki67及lncRNA H19的表达均无显著影响;而当存在一定浓度的幽门螺旋杆菌时,三黄泻心汤时处于分裂间期和前期(G0/G1期)的细胞的百分比显著的升高,处于DNA复制(S期)的细胞比例及Ki67、lncRNA H19的表达均明显的降低(P<0.05)。结论:在幽门螺旋杆菌存在时,三黄泻心汤显著抑制胃癌细胞系的增殖,可能与降低长链非编码RNA H19的表达有关。

      ABSTRACT Objective: To investigate the effect and mechanism of Sanhuang Xiexin Decoction on the proliferation of gastric cancer cells. Methods: The gastric cancer cell line HGC-27 was cultured and treated with Sanhuang Xiexin Decoction. The effect of Sanhuang Xiexin Decoction on the cell cycle of HGC-27 was detected by flow cytometry. The expression of Ki67 in HGC-27 cells was detected by immunofluorescence, and the expression of proliferation-associated non-coding RNA (H19) was detected by qRT-PCR. Results: In the absence of Helicobacter pylori, Sanhuang Xiexin Decoction had no significant effect on the cell cycle, Ki67 expression and lncRNA H19 in gastric cancer cell lines. While there was a certain concentration of H. pylori, (G0/G1 phase), the percentage of cells in DNA replica- tion (S phase) and the expression of Ki67 and lncRNA H19 were significantly decreased (P<0.05). Conclusion: Sanhuang Xiexin Decoc- tion could significantly inhibit the proliferation of gastric cancer cell lines in the presence of Helicobacter pylori, which might be related to the decrease of H19 expression.
