      阿尔茨海默病( Alzheimer's disease,AD)是一种中枢神经系统神经退行性疾病,至今尚未明确其发病机制,但基于其典型的 病理特征之一是beta淀粉样蛋白(amyloid-beta, Abeta)聚集,A茁沉积假说一直都是研究的重点。近年来,载脂蛋白E(apolipoprotein E, APOE)对Abeta代谢清除的影响备受关注。研究表明,APOE着4 等位基因是散发性AD 的危险因素,且APOE 对Abeta具有很高的亲和 力,不同亚型的APOE 对Abeta的代谢有不同的影响,这为对AD的认识、防止及治疗提供了新的研究方向。
英文摘要:
      Alzheimer's disease (AD) is a neurodegenerative disease of the central nervous system. Its pathogenesis is still incompletely understood. The accumulation of amyloid-beta (Abeta) is one of the typical pathologic symptoms, and the "amyloid hypothesis" has dominated Alzheimer research for a long time. Recently, the effect of apolipoprotein E (APOE) in Abeta metabolism attracts extensive concern. It suggests that ε4 allele is a risk factor in sporadic Alzheimer disease. Apolipoprotein E has a high affinity with A-beta, while different isoforms of apolipoprotein E have different influences on A-beta metabolism. These results provide a new way for the identification, prevention and therapy of AD.
