我院收治的47 例湿热痹阻型类风湿关节炎患者(湿热痹阻组)、43 例寒湿痹阻型类风湿关节炎患者(寒湿痹阻组)以及同期我院
体检科确诊的50 例健康体检者(对照组)作为研究对象。采用酶联免疫吸附试验(ELISA)双抗体夹心法检测所有研究对象的血清
基质金属蛋白酶-3(MMP-3)、金属蛋白酶抑制剂-1(TIMP-1)、肿瘤坏死因子-alpha(TNF-alpha)、白介素-1beta(IL-1茁),同时采用流式细胞
仪检测单核细胞表面CD147 分子的表达情况。结果:3 组研究对象血清MMP-3、TIMP-1、TNF-alpha、IL-1beta相比差异有统计学意义
(P<0.05),对照组血清MMP-3 显著低于湿热痹阻组,TIMP-1、TNF-alpha、IL-1beta则显著低于湿热痹阻组与寒湿痹阻组(P<0.05),寒
湿痹阻组血清MMP-3、TIMP-1 显著低于湿热痹阻组(P<0.05),湿热痹阻组与寒湿痹阻组血清TNF-alpha、IL-1beta相比差异无统计学
意义(P>0.05)。结论:湿热痹阻型与寒湿痹阻型类风湿关节炎患者的血清MMP-3、TIMP-1 及单核细胞表面CD147 存在着显著区别。 |

Objective:To compare and analyze the difference of arthritis related cytokines between damp heat blockage type and
cold-dampness blockage type in the patients with rheumatoid arthritis.Methods:47 cases with rheumatoid arthritis of damp heat blockage
type (damp heat blockage type group), and 43 cases with rheumatoid arthritis of cold-dampness blockage type (cold-dampness blockage
group) and 50 cases of healthy person diagnosed by physical examination (control group) were selected as research objects. The level of
serum matrix metalloproteinase3 (MMP-3) and tissue inhibitor of metalloproteinase1 (TIMP-1), tumor necrosis factor alpha (TNF-alpha)
and interleukin-1 beta (IL-1beta) was examined by enzyme linked immunosorbent assay (ELISA) double antibody sandwich method, and
the expression of mononuclear cell surface CD147 was detected using the flow cytometry assay.Results:The difference of serum level of
MMP-3, TIMP-1, TNF-琢and IL-1茁among three groups was statistically significant (P<0.05). The serumlevel of MMP-3 in control group
was significantly lower than that of damp heat blockage group, and the level of TIMP-1, TNF-琢and IL-1茁was significantly lower than
that of damp heat blockage group and cold-dampness blockage group (P<0.05), and the serum level of MMP-3 and TIMP-1 was
significantly lower in cold-dampness blockage group than that of damp heat blockage group (P<0.05). The serum levels of TNF-alpha and
IL-1beta had no significantly statistical difference between damp heat blockage group and cold-dampness blockage group (P>0.05).Conclusion:The damp heat blockage type and cold-dampness blockage type rheumatoid arthritic inflammation in patients with serum
MMP-3, TIMP-1 and monocyte surface CD147 exist significant difference. |
