目的:探讨氧化苦参碱(OMT)对大鼠缺血再灌注心肌损伤(MIRI)的保护机制。方法:随机将60 只成年Wistar 大鼠分成对
照组、MIRI 组和OMT 组,每组20 只,除对照组外,其他两组结扎30 min 后松解结扎线灌注60 min。结扎前10 min,OMT 组股静
脉输入苦参注射液120 mg/kg,对照组、MIRI组则输入等容量生理盐水。造模后,记录两组心率(HR)、左心室收缩压(LVSP)、左室
内压最大上升或下降速率(+dp/dtmax或-dp/dtmin)及血清乳酸脱氢酶(LDH),检测两组心肌组织中一氧化氮(NO)、丙二醛(MDA)、
一氧化氮合酶(NOS)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)水平。结果:与MIRI组相比,OMT 组HR、LVSP 和
+dp/dtmax、-dp/dtmin均显著升高(P<0.05),且OMT 组上述指标与对照组比较,差异均无统计学意义(均P>0.05)。与MIRI 组相比,
OMT 组的NO、NOS、SOD、GSH-PX 水平均显著升高,而MDA、血清LDH水平显著降低,比较差异均有统计学意义(均P<0.05),
且OMT 组上述指标与对照组比较,差异均无统计学意义(均P>0.05)。结论:OMT 对MIRI大鼠具有心肌保护作用,其机制可能与

Objective:To explore the protection mechanism research on oxymatrine (OMT) for the myocardial ischemia reperfusion
injury (MIRI)rats.Methods:A total of 60 Wistar rats were divided into control groups, MIRI group and OMT group randomly, with
20 rats in each group, except control group, the other two groups were ligated for 30 min then loosed the ligation line to perfusion for 60
min, 10 min before ligation, the OMT group was injected the matrine 120 mg/kg through vein, while the control group and MIRI group
was injected the equal volume normal saline. After the model was built-up, recorded the heart rate (HR), left ventricular systolic pressure
(LVSP), maximum rise or decrease rate of left ventricular pressure (+dp/dtmax or -dp/dtmax), and the levels of serum lactate dehydrogenase
(LDH)of two groups, detected the levels of nitric oxide(NO), malondialdehyde(MDA), nitric oxide synthase(NOS), superoxide dismutase
(SOD), glutathione peroxidase(GSH-PX)in myocardial tissue of two groups.Results:Compared with MIRI group, the HR, LVSP, +dp/dtmax,
-dp/dtmin and the levels of NO, NOS, SOD, GSH-PX in myocardial tissue of OMT group obviously increased, and the levels of MDA in
myocardial tissue and serum LDH obviously decreased, the differences were statistically significant (P<0.05), while above indexes in
OMT group had no statistical difference than control group (P>0.05).Conclusion:OMT has a protective effect on the myocardium of
MIRI rats, and its mechanism may be related to increasing the activity of anti oxygen free radical, improving microcirculation and relaxing
coronary artery. |
