      目的:从足月剖腹产分娩新生儿脐带中分离出人脐带间充质干细胞(UC-MSCs),探讨其在体外促进SKOV3卵巢癌细胞凋 亡,抑制其增殖的作用。方法:新鲜人脐带洗净后剥离动静脉及脐带外膜,得到脐带Wharton's胶。采用组织块贴壁法分离、纯化得 到UC-MSCs细胞,光镜下观察UC-MSCs细胞的形态及贴壁生长情况。收集UC-MSCs 细胞培养上清,加入SKOV3 细胞共培养 后,观察不同作用时间(12 h,24 h,36 h,48 h,60 h,72 h)其体外促进SKOV3卵巢癌细胞凋亡,抑制其增殖的作用。结果:光镜下 UC-MSCs 细胞成长梭状,单核,并成放射或漩涡状排列。PI染色提示,随着UC-MSCs 细胞培养上清对SKOV3 卵巢癌细胞作用时 间的增加,其发生凋亡的细胞数量增多,且具有统计学意义(P<0.05)。MTT 实验提示SKOV3 细胞增殖活力随UC-MSCs 细胞培养 上清作用时间的增加而显著下降(P<0.05),共培养24 h,48 h,72 h的抑制率分别为17.08 %,35.36 %,46.83 %。结论:UC-MSCs 在 体外具有明显促进SKOV3卵巢癌细胞凋亡,抑制其增殖的作用。

      Objective:The aim of this study is to isolate human umbilical cord-derived mesenchymal stem cells from newborn human umbilical cord and explore the effect of UC-MSCs on the proliferation, as well as apoptosis of SKOV3 cells in vitro.Methods:The fresh human umbilical cord tissues were obtained and washed under sterile conditions. Arteries and vein were removed in order to get Wharton's jelly. UC-MSCs were isolated and amplified via tissue-cultivation method. Morphology and adherent condition of UC-MSCs were observed under the optical microscope. UC-MSCs cell culture supernatant was collected, and then co-cultured with SKOV3 cells. According to co-culture time, we divided the study group into 6 groups: 12 h, 24 h, 36 h, 48 h, 60 h and 72 h. We evaluated the activities of UC-MSCs in apoptosis and proliferation of SKOV3 cells.Results:Fibroblast-like cells were successfully isolated from human umbilical cord tissues using the direct tissue-cultivation method. The UC-MSCs cells formed parallel or whirlpool-like arrays when a confluent monolayer had developed. PI stain test showed UC-MSCs could dramatically induce apoptosis of SKOV3 cells (P<0.05). MTT test revealed UC-MSCs could significantly inhibit proliferation of SKOV3 cells (P<0.05).Conclusion:UC-MSCs can be successfully isolated from newborn human umbilical cord tissue by tissue-cultivation method. UC-MSCs can induce apoptosis and inhibit proliferation of SKOV3 cells in vitro.
