      目的:近年来许多报道表明,miRNA 与一些肿瘤的发病息息相关,其其表达失调直接或间接的影响肿瘤的进展。检测 mir-26a 在子宫肌瘤组织和肌层组织中的表达,并进一步探究其对子宫肌瘤细胞周期和增殖的影响。方法:收集第二军医大学长海 医院妇产科2009 年12 月至2011 年10 月手术治疗并经病理检查确诊为子宫肌瘤的患者肌瘤组织和成对肌层组织标本13 例, Realtime PCR检测这13 例成对组织中mir-26a 的表达情况。建立可稳定传代的子宫肌瘤平滑肌细胞系后,将mir-26a转入肌瘤细 胞中使其高表达,流式细胞仪检测高表达mir-26a 后子宫肌瘤细胞周期的变化,同时用cck-8 方法验证细胞增殖活性。结果: Realtime PCR结果显示,与相应肌层组织相比,肌瘤组织mir-26a 的表达均显著降低(P<0.05)。流式检测结果显示,24 h时,与对照 组相比,mir-26a 高表达的肌瘤细胞G1 期比例增高。增殖实验显示种植细胞第二天开始,mir-26a 高表达的子宫肌瘤细胞增殖速 度显著降低。结论:与正常子宫肌层组织相比,mir-26a 在子宫肌瘤组织中表达下调,这种表达失调直接影响肌瘤细胞的增殖速度 和周期比例,提示mir-26a 在子宫肌瘤的发生发展过程中发挥了重要的调控作用。

      Objective:In recent years, many reports indicate that miRNA is closely related to the progress of tumors directly or indirectly. The study aims to investigate the expression of mir-26a in Leiomyoma uteri tissues and myometrium tissues of leiomyoma uteri and to study the effect of mir-26a on cell cycle and cell proliferation of Leiomyoma uteri tissues.Methods:13 pairs of leiomyoma uteri tissues and myometrium tissues of leiomyoma uteri patients, who have received surgical treatment and pathological examination, were collected during December 2009 to October 2011 at Gynaecology department of Changhai Hospital of The Second Military Medical University. The expression of mir-26a in leiomyoma uteri tissues and the paired myometrium tissues were detected respectively by Realtime PCR. Leiomyoma uteri cells were primary cultured fromleiomyoma uteri tissues to get a stable passaged smooth muscle cells of uterine fibroids. Cell cycle and cell proliferation activity was detected after mir-26a was over-expressed in uterine leiomyoma cells by FACS and by CCK-8, respectively.Results:Realtime PCR results showed that compared with the corresponding myometrium, the mir-26a level of leiomyoma uteri tissues was obviously down-regulated in 13 pairs of samples (P<0.05). Flow cytometry showed that, the G1 phase ratio increased at 24 h, compared with the control group. Proliferation experiments observed that proliferation rate of mir-26a high expression uterine leiomyoma cell was significantly reduced after the second day of seeding cells.Conclusion:Compared with normal myometrium, mir-26a downregulated in uterine leiomyoma. The abnormal expression of mir-26a directly affects the proliferation rate and cell cycle of uterine fibroids. This suggested that mir-26a played an important role in development of uterine leiomyoma.
