组织和癌旁组织中的lncRNA91H表达,设置干扰序列,噻唑蓝(MTT)测定细胞增殖率和流式细胞仪检测细胞凋亡率,COX 分析
lncRNA91H和预后关系。结果:癌组织lncRNA91H相对表达量为(1.83± 0.14),癌旁组织为(0.36± 0.07),差异具有统计学意义
(P<0.05);lncRNA91H 表达在不同TNM 分期、肿瘤转移以及浸润深度间差异均具有统计学意义(P<0.05)。多因素COX分析显
示,TNM 分期、肿瘤转移、浸润深度、lncRNA91H表达是影响CRC预后独立危险因素(P<0.05)。干扰序列作用于lncRNA91H表
达后,培养24 h、48 h、96 h时lncRNA91H表达组吸光度(OD)均低于对照组,且随着培养时间的延长,两组OD均显著上升(P<0.
05);阴性对照组细胞凋亡率为(5.67± 1.23)%,转染lncRNA91H 细胞凋亡率为(25.37± 0.89)%,差异具有统计学意义(P<0.05)。
结论:lncRNA91H 可以作为CRC 患者预后特异指标,并且与TNM分期、肿瘤转移以及浸润深度间等临床病理特征密切相关。 |

Objective:To investigate the expression of long noncoding RNA (lncRNA) 91H and its correlation with clinical
pathological features and prognosis in patients with colorectal cancer (CRC).Methods:A total of 80 specimens (including cancer tissues
and para cancer tissues) of patients with CRC, who underwent CRC radical resection in First Affiliated Hospital of Jiamusi University
fromJanuary 2010 to January 2012, were selected. The expression of lncRNA91 in cancer tissues and para cancer tissues was detected by
quantitative real-time PCR.The disturbance sequence was set; the cell proliferation rate was detected by thiazole blue (MTT) and cell
apoptosis rate was detected by flow cytometry; the correlation between lncRNA91H and prognosis was analyzed by COX.Results:The
relative expression amount of lncRNA91H in cancer tissues and para cancer tissues was (1.83± 0.14) and (0.36± 0.07) respectively, the
difference was statistically significant (P<0.05). The expressions of lncRNA91H in different TNM stage, tumor metastasis and invasion
depth were statistically different (P<0.05). Multivariate COX analysis showed that TNM stage, tumor metastasis, invasion depth and
expression of lncRNA91H were the independent risk factors of prognosis of CRC (P<0.05). The apoptosis rate of negative control group
was(5.67± 1.23)%, and the apoptosis rate of transfected lncRNA91H cells was(25.37± 0.89)%, the difference was statistically significant
(P<0.05).Conclusion:LncRNA91H can be used as a specific index of prognosis of CRC patients, which is closely correlated with the
clinical pathological features of TNMstage, tumor metastasis and invasion depth. |
