      目的:研究阿魏酸(ferulic acid,FA)在缺氧条件下对人脐静脉内皮细胞(human umbilical vein endothelial cells,HUVECs)增 殖、迁移和管腔样结构形成的影响。方法:原代培养人脐静脉内皮细胞,在缺氧实验条件下,细胞被分为7 组,即1 个对照组和6 个实验组。对照组采用1 %酒精处理,实验组用不同浓度(1× 10-8、1× 10-7、1× 10-6、1× 10-5、1× 10-4及1× 10-3mol/L)的阿魏酸处 理。分别采用MTS 法、划痕法、Matrigel 法分析不同浓度阿魏酸处理对人脐静脉内皮细胞的增殖、迁移和管腔样结构形成的影响。 结果:缺氧条件下,浓度为1× 10-6~1× 10-4 mol/L的阿魏酸处理能明显促进HUVECs 的增殖(P<0.05),以1× 10-5mol/L处理的效 果最好(P<0.01);与对照组相比,1× 10-6mol/L(P<0.05)、1× 10-5 mol/L(P<0.01)及1× 10-4mol/L(P<0.01)阿魏酸处理均能明显促进 HUVECs 横向迁移,以1× 10-5 mol/L 处理迁移的细胞数量最多;1× 10-8~1× 10-4mol/L 阿魏酸处理能不同程度地促进HUVECs 管腔样结构的形成,以1× 10-5mol/L处理形成管腔样结构的数量最多(P<0.01)。结论:阿魏酸在缺氧条件下能促进人脐静脉内皮 细胞的增殖、迁移和管腔样结构形成。

      Objective:To study the effects of ferulic acid (FA) on the proliferation, the migration and the formation of the tube-like structure of human umbilical vein endothelial cells (HUVECs) under hypoxia.Methods:The primary HUVECs were cultured under normoxia conditions (21 % O2, 5 %CO2 and 74 %N2), and the experimental conditions of hypoxia (1 % O2, 5 % CO2 and 94 % N2) were established. Under hypoxia, the cells were divided into seven groups, one control group and six experimental groups. The control group was treated with 1 % ethanol, and the experimental groups were treated with different concentrations of ferulic acid (1× 10-8, 1× 10-7, 1× 10-6, 1× 10-5, 1× 10-4 and 1× 10-3mol/L). The MTS assay was used to evaluat the effects of the proliferation activity of ferulic acid on HUVECs after 48 hours; the wound healing assay was used to detect the effects of the lateral migration ability of ferulic acid on HUVECs after 24 hours; the tube formation assay on Matrige was used to detect the effects of the tube-like structure formation ability of ferulic acid on HUVECs after 7 hours. SPSS 17.0 was used for statistical analysis. The experimental data of each group were statistically analyzed by Dunnett-t test, and P<0.05 was statistically different.Results:Under hypoxia, the concentrations of ferulic acid between 1× 10-8 to 1× 10-5 mol/L could promote the proliferationon of HUVECs in a concentration dependent manner, and the effects of 1× 10-5 mol/L were the best. Compared with the control group , the differences were statistically significant (1× 10-6 and 1× 10-4mol/L, P<0.05; 1× 10-5mol/L,p<0.01). Compared with the control group, ferulic acid could promote the migration of HUCECs, and the differences were statistically significant (1× 10-6mol/L P<0.05; 1× 10-5 mol/L and 1× 10-4mol/L, P<0.01). Ferulic acid had the ability to enhance tube-like structure formation of HUVECs, and compared with the control group, the differences were statistically significant (1× 10-4mol/L, P<0. 05; 1× 10-5 mol/L, P<0.01).Conclusion:Ferulic acid can promote the proliferation, the migration and the formation of the tube-like structure of human umbilical vein vascular endothelial cells under hypoxia.
