      目的:研究壮肝逐瘀煎对肝纤维化(HF)大鼠肝脏肝星状细胞(HSC)的凋亡因子Fas/FasL表达的影响,探讨该方抗HF的作 用机制。方法:58 只SD大鼠随机取14 只作为正常对照组,正常饲养。剩余大鼠采用CCl4复合因素法进行HF造模,实验第4 周 末随机处死造模大鼠4 只,证实HF形成。其余随机分为病理模型组、壮肝逐瘀煎组、秋水仙碱组及大黄蛰虫丸组,每组10 只。病 理模型组给予生理盐水灌胃处理,其他3 组分别予对应药物灌胃处理。治疗第6 周末,处死所有大鼠,获取肝组织,HE染色光镜 观察肝组织结构变化,免疫组化染色分析Fas、FasL分布。结果:与病理模型组大鼠比较,壮肝逐瘀煎治疗组肝小叶结构趋于正常, 纤维间隔明显变薄,肝组织Fas、FasL的表达显著表达显著减少(P<0.01)。结论:壮肝逐瘀煎能够明显改善HF大鼠肝脏的病理变 化,具有明显的抗HF作用,其作用机制可能与壮肝逐瘀煎能调控HSC 中凋亡因子Fas/FasL 的表达有关。

      Objective:To study the effects of Zhuangganzhuyu Decoction on the expression of apoptosis factors Fas/FasL in hepatic stellate cells (HSC) of hepatic fibrosis (HF)-stricken rats, and explore it's mechanism.Methods:14 out of 58 SD rats were randomly chosen as the Normal Control group and fed normally. The remaining rats were subject to HF modeling with CCl4 combining other factors; four rats were sacrificed to verify the formation of HF at the end of the fourth week of the experiment, and the others were randomly divided into four groups, namely the groups of Pathological Model, Zhuangganzhuyu Decoction, Colchicin and Dahuangzhechong Pill, with 10 rats in each group. The Pathological Model group was given normal saline by intragastric administration, and the other 3 groups were administered intragastrically with corresponding drugs. At the end of the sixth week, all rats were sacrificed to obtain the liver tissues, which were then subject to HE staining, and the structure changes were observed under a microscope and immunohistochemical staining for Fas and FasL distribution analysis.Results:Compared with those in the Pathological Model group, the rats in the Zhuangganzhuyu Decoction group had more normal hepatic lobules, thinner fibrous septum, and lower expression of Fas and FasL (P<0.01).Conclusion:Zhuangganzhuyu Decoction can significantly improve pathological changes in the livers of rats with HF, and has an effective anti-HF action, which may be related to its regulation of the expression of apoptosis factors Fas/FasL in HSC.
