Objective:This study aimed to construct a transferrin modified magnetic nanoparticle, which can targeted deliver doxorubicin(
DOX) to tumor cells.Methods:The magnetic nanoparticle(DOX@MNP) was prepared by the chemical co-precipitation. The zeta
potential and particle size were detected by Laser Particle Size Analyzer. The dialysis method was used to evaluate the characteristic of
drug release of DOX lipid hybrid nanoparticle. The anti-tumor activity of DOX@MNP was studied by MTT method. The cell uptake of
DOX@MNP in A549 cells was observed with flow cytometry and confocal laser scanning microscope.Results:The drug release of
DOX@MNP exhibited an acidic sensitivity property. DOX@MNPshowed the same cytotoxicity as DOX didon A549 cells. The flow cytometry
and confocal laser scanning microscope results showed that there was no significant difference in cellular uptake between DOX
and DOX@MNP in A549 cells.Conclusion:This study constructed a magnetic nanoparticle modified with transferrin to deliver DOX to
tumor cells. DOX@MNP had the same anti-tumor activity as free DOX in vitro. DOX@MNP is worthy of investigation in vivo. |
