目的:探讨交感神经分泌的神经肽Y(NPY)和感觉神经分泌的钙基因相关肽(CGRP)在体内骨折愈合的不同阶段的变化及意
义。方法:选择6-8 月龄的雄性大鼠,建立大鼠的股骨闭合骨折模型,术后2、4、8、12 周取材。进行扫描电镜,免疫组织荧光染色和
血清Elisa 检测。结果:①骨折愈合不同时期感觉神经肽类物质CGRP 和交感神经肽类物质NPY 都有表达,且其含量有先增加后
减少的趋势,并在骨折后8 周含量达到最高。②骨折愈合不同阶段的大鼠血清感觉神经肽类物质CGRP 和交感神经肽类物质
NPY 均呈上升趋势,差异有统计学意义(P<0.05),且NPY 的含量比CGRP 的含量高。骨折后2-4 周,CGRP含量增加较快;骨折后
4-8 周NPY 含量增加较快。结论:骨折愈合的不同阶段,感觉神经肽类物质CGRP 和交感神经肽类物质NPY 含量先升后降,对不
同阶段的骨形成及骨吸收产生影响。 |

Objective:To explore the changes and significances of the sensory nerve secretion of peptide CGRP and the sympathetic
nerve secretion of peptide NPY in the different fracture healing stages of rats with closed femur fracture.Methods:6-8 months old
male rats were chosen and established a closed fracture model. At 2, 4, 8and 12 weeks after surgery, samples were harvested for examinations
of histology by scanning electron microscopy (SEM), immunofluorescence staining and ELISA.Results:The sensory neuropeptide
substance CGRP and sympathetic neuropeptide substance NPY were expressed in the different stages of fracture healing. The content of
them increased first and then decreased, reached the highest level at the 8 week after fracture. In the different stages of fracture healing,
the sense of neuropeptide substance CGRP and sympathetic neuropeptide substance NPY showed an upward trend in the serum of rats
(mean± SD, p<0.0001), and the content of NPY was higher than CGRP. During 2-4 week after fracture, the content of CGRP increased
faster; while NPY increased faster during 4-8 week after fracture.Conclusion:The content of CGRP and sympathetic neuropeptide substance
NPY were different in fracture healing process, making effects on bone formation and bone resorption, which promotes the healing
of fracture. |
