      目的:探讨熊去氧胆酸对肝硬化大鼠血清IL-6、MMP-3、TIMP-1 水平及胃肠Cajal细胞c-kit 表达的影响。方法:选取72 只大 鼠,随机分为四组,每组18 只。空白组给予正常饲养,其余三组均注射CCl4,给予酒精和高蛋白饮食,熊去氧胆酸组大鼠注射熊去 氧胆酸1.5 mg/Kg·d;吗丁啉组给予吗丁啉0.85 mg/Kg·d;模型组仅给予注射生理盐水。治疗结束后,对比各组血清IL-6、MMP-3、 TIMP-1 水平及胃肠Cajal细胞c-kit含量。结果:与空白组比较,模型组大鼠血清IL- 6、MMP-3、TIMP-1 水平均明显升高(P<0.01), 胃肠Cajal细胞c-kit含量显著降低(P<0.01)。与模型组相比,熊去氧胆酸组与吗丁啉组大鼠血清IL-6、MMP-3、TIMP-1 水平均显著 降低(P<0.01),小肠推进率增高(P<0.01),胃肠Cajal 细胞c-kit 含量显著升高(P<0.01);与吗丁啉组相比,熊去氧胆酸组血清IL- 6、 MMP-3、TIMP-1 水平较低(P<0.01),小肠推进率、胃肠Cajal细胞c-kit 含量显著升高(P<0.01)。结论:熊去氧胆酸对肝硬化具有较好 疗效,可改善肝纤维化和胃肠功能,这可能与其降低血清IL-6、MMP-3、TIMP-1 水平及提高胃肠Cajal细胞C-kit 含量有关。

      Objective:To investigate the effect of ursodesoxycholic acid on the IL-6, MMP-3, and TIMP-1 in serum and c-kit expression in Cajal cells of rats with liver cirrhosis.Methods:72 rats were selected and randomly divided into four groups. The blank group were get normal feeding and others group were treated with CCl4 alcohol and high-protein diet. The ursodesoxycholic acid group was treated with 1.5 mg/Kg·d ursodesoxycholic acid. The domperidone group was treated with 0.85 mg/Kg·d domperidone. The model group was treated with physiological saline. Then the serumlevels of MMP-3, TIMP-1 and IL-6 and expression of c-kit in gastrointestinal Cajal cells of different groups were observed and compared.Results:Compared with the blank group, the serum levels of IL-6, MMP-3, TIMP-1 were higher (P<0.01), the C-kit expression in gastrointestinal Cajal cells was lower (P<0.01). Compared with the model group, the serum IL-6, MMP-3, TIMP-1 levels were lower in the ursodesoxycholic acid group and the domperidone group (P<0.01), the intestinal propulsion rate and the C-kit expression in gastrointestinal Cajal cells were higher(P<0.01).Conclusion:Ursodesoxycholic acid had good curative effect in the treatment of liver cirrhosis, which could improve the liver fibrosis and gastrointestinal function.
