目的:探讨不同睡眠剥夺时间对大鼠认知功能的影响以及对下丘脑内单胺类神经递质去甲肾上腺素、多巴胺、五羟吲哚乙
酸、五羟色胺的含量的影响。方法:32 只健康雄性wistar大鼠随机分为4 组,即96 h、120 h、144 h 睡眠剥夺,正常对照组。利用睡眠
剥夺箱建立大鼠SD 模型,避暗穿梭法测试大鼠认知功能,高效液相电化学检测法测定下丘脑内单胺类神经递质含量。结果:大鼠
避暗穿梭实验,与对照组比较,96 h、120 h组大鼠潜伏期显著缩短(P<0.05);与对照组比较,各组大鼠下丘脑内NA 含量均有下降
(P<0.05);与对照组比较,各组大鼠下丘脑内DA含量均显著下降,(P < 0.01),96 h、120 h、144 h组间比较,表现出含量逐渐减少
的趋势;与对照组比较,各组5-HIAA含量均有上升,且120 h组明显高于其他各组(P<0.05),其他组无显著性差异(P>0.05);与
对照组比较,各组5-HT 含量均有升高,120 h、144 h组显著升高(P<0.01),96 h组无显著性(P>0.05)。结论:睡眠剥夺可以使大
鼠中枢NA、DA 含量下降,5-HIAA、5-HT 含量升高,且随着睡眠剥夺时间的延长,变化更为明显,这可能是睡眠剥夺损害认知功
能的原因之一。 |

Objective:This study aim to research the effect of different duration of sleep deprivation on cognitive function and
monoamine neurotransmitter levels in hypothalamus of rats.Methods:32 Male wistar rats were randomly divided into four groups: 96 h,
120 h, 144 h sleep deprivation, control group. SD model was established by modified multiple platform method, cognitive function was
assessed with passive avoidance experiment, monoamine neurotransmittersin hypothalamus was measured by HPLC with electrochemical
detection.Results:Incubations were significantly decreased in 96 h and 120 h groups compare with the control group (P<0.05).
Compared with the control group, the content of NA were decreased in all the other groups(P<0.05), as well as the content of DA(P<
0.01). With the increasing of sleep deprivation time, the result shows decreasing trend. The levels of 5-HIAA were decreased in all the
other groups compare with the control group, especially in 120 h group (P<0.05). Compared with control group, 5-HT increased
significantly in the SD groups.Conclusion:Sleep deprivation could decrease the content of NA and DA, and increase the content of
5-HIAA and 5-HT quite the contrary, which may be the reason of impairing cognitive function. |
