| 目的：分析不同年龄组包皮组织中表皮干细胞Integrin beta1 的表达，为组织工程学种子细胞的选用提供依据。方法：利用DispaseⅡ
和BSA 共同作用，分散表皮和真皮组织部分。对不同年龄组分离的表皮干细胞进行Integrin beta1 免疫荧光染色，利用流式细
胞仪检测不同年龄组Integrin beta1表达的阳性率。结果：不同年龄组分离得到的表皮干细胞形态一致。细胞生长状态和生长速度以
7 岁年龄组为最佳，其次是16 岁年龄组，最后是30 岁年龄组。流式细胞仪检测不同年龄组Integrin beta1 的表达，7 岁组阳性率为
93 %，16 岁组阳性率为56 %，30 岁组阳性率仅为3.5 %。结论：随着年龄的增长，皮肤组织中表皮干细胞的含量降低，低年龄段皮
肤组织是组织工程学种子细胞的良好选择。 |

| Objective:To investigate the differential expression of Integrin beta1 in epidermal stemcells among different age groups.Methods:The Epidermal stem cells were obtained by digesting human foreskin with DispaseⅡ and BSA and Trypsin. The expression of
Integrin beta 1 of epidermal stem cells was detected by Flow cytometry (FCM).Results:The epidermal stem cells marked with beta1-integrin
and sorted by flow cytometry were similar in shape and size among different age groups. The cell growth state of the group aged 7were
the best among the three groups. The rate of positive expression of beta1-integrin in the group aged 7, aged 16 and aged 30 were 93%,56%,
3.5%, respectively.Conclusion:The expression of integrin beta1 in three epidermal stem cells was decreased with the age, suggesting that
the stemcells in skin tissues are decreased with age, and the lower age will be the best choice for tissue engineering. |
