A549两种细胞经Hoechst33342 染色后,流式细胞仪405 nm激光激发检测DNA 含量,利用软件计算出处于G0/G1 期、S 期和
G2/M期细胞的百分比,以PI染色法结果作为对照。结果:SW480 和A549细胞经Hoechst33342 染色后各期的细胞百分比与PI染
色法基本一致,无明显差异(P>0.05)。结论:405 nm激光激发Hoechst33342 染色细胞DNA 结果可靠,可作为紫外检测的替代方

Objective:To observe the effect and influential factors of flow cytometry with 405 nm laser using Hoechst33342 in
detection of DNA content.Methods:SW480 and A549 cells were stained with Hoechst33342 and then excited by 405 nm laser to detect
DNA content, percentages of cells in G0/G1, S and G2/Mphases were calculated by software,results of PI staining method were used for
control.Results:The percentages of SW480 and A549 cells in each period stained with Hoechst33342 were basically same with PI
staining method(P>0.05)Conclusion:405 nmlaser is credible in exciting Hochest33342 staining cell DNA, so it can replace ultraviolet
laser to detect DNA content. |
