述了NAD+的生物合成、其参与的信号转导过程等研究的最新进展以及它与细胞增殖动态平衡的联系,以及基于此的一些抗癌药

NAD+ is a key molecule in living organisms. The processes of energy metabolism and cell signal transduction have
been linked by NAD+. NAD+ is required for ADP ribosylation and NAD+-dependent protein deacetylation. The NAD+-dependent signal
pathways regulate significant events such as transcription, cell cycle progression, DNA repair and apoptosis. Many of the processes relate
to the cancer development. Given the research of NAD+-dependent signal pathways, the strategy of targeting NAD+ metabolism for
cancer treatment has been developed. This review explored most recent advances in studies of NAD+ biosynthesis, NAD+-dependent
signal pathways and the intimate relationship to the maintenance of cell proliferation. These studies also give the clue of exploitation of
the new anticancer agents. |
