目的:通过建立高血脂动物模型,探索高血脂动物血浆丁酰胆碱酯酶(BuChE)的活性与血脂指标的相关性,为BuChE 活性
作为高血脂疾病标志物的可能性提供参考。方法:雄性新西兰兔14 只,随机分为两组(每组各7 只),即普通饲料组(C 组)和高胆
固醇饲料组(H 组)。连续喂养12 周,每二至三周称重采血,采用Ellman 法检测BuChE活性,按照试剂盒使用说明测定血浆总胆
固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C);12 周后,麻醉兔子开胸采集胸主动脉,
经10 %的中性福尔马林固定后进行苏丹Ⅳ脂肪染色。结果:兔子喂食高胆固醇饲料12 周后,H组动脉出现斑块,C 组无斑块形
成;兔子在食用高胆固醇饲料的过程中,血浆TC、LDL-C、HDL-C显著升高(P<0.01),而BuChE 活性无显著变化(P>0.05);分析正
常兔子和高血脂兔子血浆BuChE活性与TC、TG、HDL-C、LDL-C 的皮尔森相关系数,发现均无相关性(r<<1,P>0.05)。结论:本项
研究未在正常兔子和高血脂的兔子中发现BuChE 与TC、TG、HDL-C、LDL-C 存在相关性,说明通过喂食兔子高胆固醇饲料建立

Objective:To compare the correlation between butyrylcholinesterase (BuChE) activity and blood lipid in normal and
hyperlipidemia rabbit and to explore the potential of using BuChE as a biomarker for hyperlipidemia.Methods:14 male New Zealand
rabbits were randomly divided into two groups (7 rabbits each), normal diet group (group C) and high-cholesterol diet group (group H).
All rabbits were fed with either normal diet or high-cholesterol diet for 12 weeks. The body weight of each rabbit was recorded and the
peripheral blood was collected every 2 to 3 weeks. The activity of BuChE was detected with Ellman's assay and the concentration of plasma
total cholesterol (TC), triglyceride (TG), high-density lipoprotein cholesterol (HDL-C) and low-density lipoprotein cholesterol
(LDL-C) was analyzed with commercial kits. After 12 weeks, rabbits were anaesthetized with urethane, the aorta thoracicas were taken
out and fixed in 10 % neutral formaldehyde for 24 hours, then stained with SudanⅣ .Results:After 12 weeks, clear atherosclerotic
plaques were found in the aorta thoracicas of high-cholesterol diet group (Group H) but not in that of normal diet group (Group C). With
the progress of high cholesterol diet, the TC, LDL-C and HDL-C level in the plasma of group H was much higher compare to that in
groupC (P<0.01), while BuChE activity remained the same compare to the 0 week. The results of the Pearson correlation indicated that
there was no significant correlation between BuChE activity and TC, TG, HDL-C, LDL-C level (r<<1, P>0.05).Conclusion:The result
of this research showed that there was no significant relationship between BuChE activity and TC, TG, HDL-C, LDL-C level, which suggests
that the cholesterol-fed hyperlipidemia rabbit model is not suitable for the correlation analyses between BuChE activity and TC, TG,
HDL-C and LDL-C. |
