目的:探讨Bmi-1 基因表达在骨髓增生异常综合征(MDS)中的临床意义。方法:选择2011 年1 月~2012 年12 月我院收治的
MDS患者41 例为研究组,另选取非恶性血液病患者20 例为对照组,检测Bmi-1 的表达水平,并检测其骨髓白血病干细胞免疫表
型(CD34+CD38-CD123+),然后分析患者Bmi-1 的表达水平与骨髓原始细胞比例及骨髓染色体核型的关系。结果:Bmi-1表达水平
研究组患者明显高于对照组,且研究组中RA 患者明显低于RAEB 患者,但RA 患者及RAEB 患者均高于对照组(P<0.01);
Bmi-1 基因高表达组白血病干细胞免疫表型CD34+CD38-CD123+/CD34+明显高于Bmi-1 基因低表达组患者(P<0.01);Bmi-1基因
高表达组中,骨髓原始细胞>5%的病例数及染色体不良核型发生率均高于低表达组。结论:Bmi-1 基因的表达可以作为MDS 患
者在分子水平上的恶性程度标志之一。 |

Objective:To explore the clinical significance of Bmi-1 gene expression in myelodysplastic syndrome (MDS). Methods:41 cases of MDS patients who were treated in our hospital were selected from January 2011 to December 2012 as the study group, and
20 non-malignant hematologic patients as control group, then detected the level of Bmi-1 gene expression in bone marrow mononuclear
cell and immunophenotype CD34+CD38-CD123+ in leukemia stem cells of MDS patients, to analyze the relation between Bmi-1 gene expression
level and proportion of bone marrow original cell and bone marrow karyotype.Results:Bmi-1 gene expression level in bone
marrowmononuclear cells of research group was significantly higher than control group (P<0.05), and the patients of RAwas significantly
lower than the patients of RAEB in research group, but both patients were significantly higher than control group (P<0.01); The proportion
of CD34+CD38-CD123+/CD34+ in high Bmi-1 gene expression level group was significantly higher than the low Bmi-1 gene expression
level group (P<0.01), and the patients with bone marrow original cell>5% and The incidence of abnormal chromosome in high
Bmi-1 gene expression level group was significantly higher than the low Bmi-1 gene expression level group.Conclusion:Bmi-1 gene expression
level can be regarded as a sign of the molecular malignancy of MDS. |
