      目的:从基因表达谱的角度探究活血化瘀方抗糖尿病心肌病的药理调控机制。方法:采用链脲霉素建立糖尿病心肌病大鼠 模型后,随机分组并对其中一组给予活血化瘀方灌胃,检测生理数据,并取样进行基因表达水平检测。针对芯片数据,初处理后依 次进行:基因表达差异分析、基因表达与性状相关性分析、基因连接性差异分析和加权的基因共表达网络分析,最后整合上述多 种分析结果,寻找关键基因。结果:成功建立糖尿病心肌病大鼠模型,并得到一系列从不同角度分析的结果:一千多个差异表达基 因,五千多个差异连接性基因,鉴定出三个共表达网络中的关键模块以及三个关键基因。结论:活血化瘀方通过调节与糖尿病心 肌病或者糖尿病密切相关的生物分子及其活动,同时综合作用于多种基础的细胞生理活动,从而影响糖尿病心肌病的发展。
英文摘要:
      Objective:To explore the pharmacological mechanism of traditional Chinese medicine (TCM) Huoxuehuayu prescriptions against diabetic cardiomyopathy on molecular level through gene expression profiles.Methods:The rats were randomly divided into model group and traditional Chinese medicine group. Diabetic cardiomyopathy model was induced by streptozotocin (STZ). After that, the rats in the TCM group were administrated with Huoxuehuayu prescriptions. Microarray data and some physiological data were collected from these two groups of samples. After raw-data processing, the study combined different approaches to comprehensively interpret the data: identify genes showing differential expression, or different connectivity, or strongly correlated to physiological character; construct weighted gene co-expression networks and determine significant modules, and the key players. Gene-set enrichment analysis, pathway mapping were also done.Results:The study successfully established a rat model of diabetic cardiomyopathy and obtained a range of results from different perspectives: more than one thousand differentially expressed genes; over five thousand genes with different connectivity; three key modules in the co-expression network, as well as three crucial genes.Conclusion:The TCM Huoxuehuayu prescriptions affect the development of diabetic cardiomyopathy by regulating some molecules and activities closely associated with the disease or with diabetes, as well as a variety of basic physiological activities in myocardial cells.
