      神经系统变性疾病(ND)的病理特征是蛋白质聚集在细胞内或细胞外形成异常的堆积体而导致神经毒性。双分子荧光互补 技术(BiFC)是研究ND 相关蛋白质寡聚化分子机制的重要工具。与其他技术不同的是,BiFC 不但可以在生理环境中对蛋白质- 蛋 白质之间的相互作用进行可视化检测,而且能够提供详细的亚细胞定位信息。本文通过综述BiFC技术的原理、优缺点及其在ND 研究领域中的应用进展,探讨该技术对揭示寡聚体和包涵体形成原因方面的潜力,为神经系统疾病开发新的治疗策略提供重要 参考。
英文摘要:
      Neurodegenerative diseases (ND) are characterized by the aberrant accumulation of misfolded and aggregated proteins inside or outside the cells which would lead to the neurotoxicity. It is proved that the bimolecular fluorescence complementation assay (BiFC) might be an excellent method of exploring the aberrant protein-protein interactions (PPI) that involved in ND. BiFC could provide detailed information for the subcellular localization of the PPI and can be carried out in a physiological environment which is different from other techniques. This review focused on the application and progression of BiFC in the field of ND by analyzing the formation of the characteristic oligomeric species, advantages and disadvantages and inclusion bodies in order to make some references for the treatment of ND.
