      目的:综述噬菌体抗体库技术的研究进展,介绍该技术的原理,构建,筛选和应用,为抗肿瘤抗体药物研发提供参考。方法: 采用文献综述的方法,筛选近5 年来噬菌体抗体库技术试验论文,对噬菌体抗体库技术的原理,构建,筛选和应用进行总结。结 果:噬菌体抗体库主要分为免疫抗体库和非免疫抗体库两大类;噬菌体抗体库筛选技术包括亲和筛选、细胞筛选和生物体内筛选 三种;噬菌体抗体库技术主要应用于肿瘤标志物的识别和肿瘤诊断,抗肿瘤抗体药物的筛选和制备。结论:噬菌体抗体库技术方 便、快速、高效,可以在体外环境下培养,这些特点决定了其在肿瘤标志物的发现和肿瘤抗体药物研发中的广泛应用。目前噬菌体 抗体库技术还存在一定缺陷,但技术的不断发展和革新必然使噬菌体抗体库技术成为研制抗体药物的新思路,极大促进了肿瘤 抗体药物的研发。
英文摘要:
      To summarize the principle, building, screening and applications of phage antibody library technology, and thus to provide a reference for the anti-tumor antibody drug development. The principle, building, screening and applications of phage antibody library technology are reviewed based on experimental papers using phage antibody library technology in the recent five years. Phage library is divided into two categories: immune antibody library and non-immune antibody library; the screening techniques include affinity screening, cell-based screening and in vivo screening; it is mainly used to identify tumor markers and for tumor diagnosis, screening and preparation of anti-tumor antibody drugs. Phage antibody library technology is convenient, fast, efficient and can be cultured in vitro environment, which determines its wide application in the discovery of tumor markers and anti-tumor antibody drug research and development. Currently, there are still some flaws in phage antibody library technology, but as the technology continues to evolve and be innovated, phage antibody library technology is bound to become a new way to research and develop antibody drugs, which will greatly promote the development of anti-tumor antibody drugs.
