      目的:比较A(beta)在两种阿尔茨海默病转基因小鼠模型脑新皮质区分布的差异。方法:采用18 月龄雄性APP/PSl 双转基因 (2× Tg-AD)小鼠与同龄同性别APP/PSl/tau 三转基因(3× Tg-AD)小鼠,取新皮质区脑组织行6E10 单克隆抗体免疫组化染色等方法显示A(beta)阳性神经元及斑块,观察其分布与形态等的差异,图像分析系统定量比较其量的变化。结果:在新皮质区2×Tg-AD组A(beta)阳性产物主要位于细胞外即细胞外A(beta)(eA(beta)),形成大量的老年斑,细胞内阳性产物少;而3× Tg-AD 组A(beta_阳性产物主要位于神经元细胞内即细胞内A(beta)(iA(beta)),但老年斑少见。结论:2× Tg-AD组与3× Tg-AD组A(beta)阳性产物在新皮质区分布的差异可能反映了两种AD小鼠模型神经病理等改变的不同。

      Objective:To compare the distribution and morphological features of A(beta)Immunoreactive products in the neocortex of the APP/PSl (2× Tg-AD) and APP/PSl/tau (3× Tg-AD) model of Alzheimer’s disease.Methods:18-month old mice of 2× Tg-AD and the age-matched mice of 3× Tg-AD were included in this experiment. In the neocortex, 6E10 immunohistochemical staining was applied to show the distribution and morphological features of A(beta)immunoreactive products, and image analysis system was applied to quantitatively analyze the difference of the products.Results:The deposition of A(beta)in the neocortex of 2× Tg-AD was mainly seen as extracellular A beta(eA beta), and formed a lot of positive plaques. The positive products were distributed as intracellularly A beta(iA beta) for the 3× Tg-AD, but the A beta positive plaques were rarely seen.Conclusion:The differences of distribution of A beta positive products in the neocortex between the 2× Tg-AD and 3× Tg-AD mice likely suggested the differences of neuropathological changes of the two AD models.
