      目的:证明NDV D90 是否具有抑制口腔鳞癌细胞系迁移和侵袭的能力。方法:免疫荧光染色法检测D90对细胞微管和微丝 形态的改变;Transwell 法检测D90 对HN-6 细胞迁移和侵袭率的抑制作用;蛋白印迹法检测D90 对于SP1、RECK、MMP-2 和 MMP-9 表达的影响;明胶酶谱法用于检测MMP-2 和MMP-9 活性的改变。结果:实验结果表明,D90 通过改变细胞的微管和微丝 形态来抑制细胞的能动性;D90 具有抑制HN-6 细胞的迁移和侵袭的功能。同时,D90 通过下调SP1 和上调RECK的表达来抑制 MMP-2 和MMP-9 的表达和活性。结论:NDV D90 能够有效地抑制口腔鳞癌细胞系HN-6 的迁移和侵袭,为一种新的抗肿瘤制剂 提供了临床试验基础。
英文摘要:
      Objective:To detect that whether NDV D90 has the ability to inhibit the migration and invasion of oralsquamous cell carcinoma cell line.Methods:Immunocyochemistry was used to detect the changes of microtubules and microfilaments induced by D90; the effect of D90 migration and invasion rate was assayed by Transwell assay; the change of SP1, RECK, MMP-2 and -9 expression induced by D90 was detected by western blot and the change of MMP-2 and -9 activity was detected by gelatin zymography.Results:The results demonstrated that D90 changed the morphology of microtubules and microfilaments to inhibit the cell motility; D90 possessed the ability of inhibiting the migration and invasion rate of HN-6 cells. Meanwhile, D90 significantly decreased the expression of sp1 and increased the expression of RECK to suppress the expression and activity of MMP-2 and MMP-9.Conclusion:NDV D90 possessed the ability to inhibit obviously the migration and invasion of HN-6 cell.The finding provided a basis for clinical trials of NDV D90 to be a new antitumor agents.
