      目的:检测白介素-3 受体α 亚基(IL-3Rα)在急性髓细胞性白血病(acute myeloid leukemia,AML)中的表达,研究其在AML 发病和预后评估中的意义。方法:应用逆转录- 聚合酶链反应(RT-PCR)法检测57 例AML 患者IL-3Rα 的表达,其中包括52 例 初治和复发患者(6 例M1、28 例M2、10 例M3、3 例M4、5 例M5)及5 例缓解患者,另10 例正常人作为对照。结果:①10 例正常 人和5 例AML 缓解患者未检测到IL-3Rα 表达。②52 例初治和复发的AML 患者中IL-3Rα 阳性表达21 例,占40.38%,M1~M5 各型患者的阳性率差异无显著性(P>0.05)。初治和复发AML 患者之间IL-3Rα 阳性表达无明显差异(P>0.05)。③AML 患者的 IL-3Rα 阳性表达与外周血白细胞计数、骨髓中原始细胞比例、CD34 阳性表达有关(P<0.05)。④AML 患者中IL-3Rα 阳性表达的 患者CR 率低于阴性者(P<0.05)。结论:①IL-3Rα 在AML 发病中有一定作用。②IL-3Rα 基因可作为AML 预后不良的一个指 标。

      Objective: To investigate the expression and significance of IL-3R α in 57 paitients with acute myeloid leukemia (AML). Methods: RT-PCR method was perfomed to examine the expression of IL-3Rα in bone marrow or peripheral blood mononuclear cells from 57 paitients with AML. The paitients included 52 newly diagnosed and relapse paitients (6 M1, 28 M2, 10 M3, 3 M4, 5 M5) and 5 CR patients. 10 healthy subjects were used as control group. Results: ①5 CR patients didn't express IL-3Rα mRNA.② The percentage of IL-3Rα mRNA expression in AML was 40.38%. IL-3Rα mRNA was expressed in all AML subtypes, and there was no difference among AML subtypes. In AML paitients, there was no difference between newly diagnosed and relapse patients. ③In AML patients, the expression of IL-3Rα mRNA related to white blood count, blast cells ratio and CD34 positive expression(P<0.05).④The CR rate of AML patients who expressed IL-3Rα was significantiy lower than that of who didn't express IL-3Rα (P<0.05). Conclusions: ① IL-3Rα gene might play an important role in the pathogenesis of AML.②IL-3Rα gene may be a poor prognostic factor in AML.
