      目的:研究L- 精氨酸和雨蛙素分别诱导SD 大鼠急性胰腺炎(AP)模型的差异,为进一步研究急性胰腺炎提供可靠模型。方 法:L- 精氨酸采用3 次腹腔注射,间隔1 h,雨蛙素采用7 次腹腔注射,间隔1 h 诱导急性胰腺炎模型。碘- 淀粉比色法检测血清 淀粉酶水平,血清脂肪酶测定试剂盒检测脂肪酶活性,胰腺组织切片观察组织的破坏情况,TUNEL 法检测腺泡细胞凋亡。结果: ①L- 精氨酸诱导的大鼠模型血清淀粉酶和脂肪酶水平在诱导成功后6 h 即显著升高,蛙皮素诱导的大鼠模型在12 h 显著升高, 与正常对照组比较均有统计学差异(P<0.05),提示急性胰腺炎建模成功。②L- 精氨酸诱导的模型中胰腺组织结构破坏,有大片出 血坏死灶、大量炎细胞浸润;而蛙皮素诱导的模型组织腺泡、间质水肿,炎性细胞浸润,少量散在出血坏死灶,血管变化常不明显, 渗液清亮。结论:L- 精氨酸和雨蛙素均能诱导SD 大鼠急性胰腺炎模型,L- 精氨酸诱导重症急性胰腺炎,雨蛙素诱导轻型急性胰 腺炎,是研究急性胰腺炎的良好模型。

      Objective: To investigate the difference of the models of acute pancreatitis(AP) induced by L-arginine and cerulein, so as to provide reliable models for further study of the acute pancreatitis. Method: Acute pancreatitis was induced in Sprague-Dawley rats either by giving three intraperitoneal injections of L-arginine, each at concentrations of 200 mg/100g body weight, with an interval of 1 h, or by giving seven intraperitoneal injections of cerulein, each at concentrations of 40 μg/kg body weight, with an interval of 1 h. Severity of AP was assessed by analysis of serum amylase and lipase activity. Samples of pancreas were taken for measuring hematoxylin-eosin staining. Apoptosis in pancreatic acinar cell was determined with terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate nick-end labeling technique (TUNEL). Result : ①Serum amylase and lipase activity has significantly increased at 6 h timepoint in the model of AP induced by L-arginine, and more higher with the extension of time. It has significantly increased at 12 h timepoint in the model induced by cerulein. Both have significantly difference(P<0.05). ②The administration of L-arginine caused severe necrotizing pancreatitis, including large hemorrhage focal necrosis, and obvious inflammatory infiltration. However, there were edema of acinus and interstitial, a small amount of inflammatory cell infiltration in the model, which induced by cerulein. Conclusion : Acute pancreatitis were induced by L-arginine and cerulein. L-arginine could induce severely, and cerulein induce mildly. There both are feasible models of acute pancreatits for further research.
