      目的:建立检测HSF1 mRNA 的real-time PCR 的方法,了解肝细胞癌患者外周血中HSF1 的表达水平及其与各临床病理特 征之间的关系。方法:利用real-time PCR 的方法检测20 例肝细胞癌患者及20 例正常人群外周血中HSF1 mRNA 的表达量。结 果:肝细胞癌患者外周血中的HSF1 mRNA 表达量显著高于正常人群(P<0.05);肝细胞癌患者外周血中HSF1 mRNA 的表达水 平在不同性别、肿瘤大小、门静脉侵犯情况、HbsAg 水平及AFP 水平的患者中的差异无统计学意义(P>0.05);在不同病理分化程 度、TNM 分期的患者中的差异有统计学意义(P<0.05)。结论:real-time PCR 技术可以成功检测外周血中HSF1 mRNA 的表达量, HSF1 可能与肝细胞癌的发生发展密切相关。
英文摘要:
      Objective: To establish a quantitative real-time PCR method for detection of HSF1 gene expression level in human hepatocellular carcinoma and to investigate the relationship between HSF1 mRNA expression level and the clinicopathological characters. Methods: Real-time PCR was employed to determine the expression level of HSF1 mRNA in peripheral blood of 20 specimens of human hepatocellular carcinoma patients and normal human. Results: Comparing with normal human, HSF1 present higher expressions in human hepatocellular carcinoma patients (P<0.05). There is no statistical differences of HSF1 mRNA expression level between the group with different sexes, tumour diameters, portal involvement levels, HBsAg levels and AFP levels (P>0.05). There is statistical differences of HSF1 mRNA expression level between the group with different differentiations and TNM stages (P>0.05). Conclusion: The established quantitative real-time PCR method can successfully detect the expression level of HSF1 mRNA, which may be closely related to the occurrence and development of human hepatocellular carcinoma.
