目的:探讨肌肽对高糖环境下心肌成纤维细胞中胶原生成的影响及作用机制。方法:原代培养心肌成纤维细胞,将细胞分
为正常糖组( NG, 5.5mmol/L glucose )、高糖组( HG, 25mmol/L glucose )、高糖+10mmol/L 肌肽组、高糖+20mmol/L 肌肽组、高糖
+40mmol/L 肌肽组、高糖+SB 组( HG+10 μmol/L SB203580 )、高糖+PD 组( HG+10 μmol/L PD98059 )。ELISA 检测胶原Ⅰ、Ⅲ的
含量,Western blot 检测TGF-β1、p-p38 MAPK 和p-ERK(1/2)等蛋白的表达。结果:与正常糖组相比,高糖组中胶原Ⅰ和胶原Ⅲ含
量增加( P<0.01 );TGF-β1 、p-p38 和p-ERK 等表达增加( P<0.01 ) ; 与高糖组相比,高糖+ 肌肽组中胶原Ⅰ、Ⅲ、TGF-β1 、p-p38、和
p-ERK 等均降低( P<0.05 );高糖+SB 组和高糖+PD 组中胶原Ⅰ、Ⅲ表达减少( P<0.05 )。结论:肌肽对心肌成纤维细胞中胶原生

Objective: To investigate the effect and mechanism of carnosine on collagen content in rat cardiac fibroblasts under
high glucose condition. Methods: The rat cardiac fibroblasts were divided into seven groups, NG group (5.5mmol/L glucose), HG group
(25mmol/L glucose), HG+10mmol/L carnosine group, HG+20mmol/L carnosine group, HG+40mmol/L carnosine group, HG+SB group
(HG+10 μmol/LSB203580 ), HG+PD group (HG+10 μmol/L PD98059 ). The collagenⅠand Ⅲ contents were detected by ELISA , and
Western blotting was used to detect the protein expression level of TGF-β1, p-p38 MAPK and p-ERK (1/2). Results: By the end of the
experiment, compared with NG group, the expression level of collagen Ⅰand collagen Ⅲ were significantly higher in the HG group
(P<0.01); The protein expression level of TGF-β1, p-p38, p-ERK were also significantly higher in the HG group (P<0.01). However,
after supplement the carnosine, the level of collagen Ⅰ, collagen Ⅲ, TGF-β1, p-p38 and p-ERK were significantly reduced in HG+Car
group than HG group (P<0.05); Compared with HG group, the collagen Ⅰand Ⅲ were also significantly decreased in HG+SB group and
HG+PD group. Conclusion: Carnosine reduced collagen production under high glucose via MAPK signaling pathway. |
