目的:观察电磁脉冲Electromagnetic Pulse(EMP)对C57BL/6J 小鼠胸腺的影响。方法:50 只C57BL/6J 小鼠按体重区组随机
化分为对照组和辐照组,每组25 只。EMP 每天照射400 次,连续照射7 天,照后1d(天)、3d、7d、14d、28d 共5 个时间点杀取胸腺。
2 只辐照组和2 只对照组杀取的胸腺做HE 染色,观察其病理改变;3 只辐照与3 只对照组的小鼠,杀后称取小鼠的体重和胸腺的
重量,计算胸腺指数;然后提取T 淋巴细胞进行计数;同时取小鼠外周血检测其中的IL-4 的水平。结果:照后1d 胸腺的切片没有
明显改变。7 天后,胸腺开始有出血,结构不清;胸腺指数呈现递减的趋势,但辐照组与对照组没有的差异没有统计学意义;T 淋巴
细胞数的变化也呈现先减后增的趋势,在第1d、14d、28d 辐照组与对照组的差异没有统计学意义,在第3d 和7d 辐照组的细胞数
小于对照组的细胞数(P<0.05);辐照组与对照组的外周血IL-4 水平的差异也没有统计学意义。结论:电磁脉冲对雄性Balb/c 小
鼠胸腺结构造成一定的损伤,但胸腺指数改变不显著,T 淋巴细胞数量增加。表明EMP 对胸腺有一定的作用,但是胸腺不是EMP
作用的敏感器官。 |

Objective: To investigate the effect of electromanetic pulse on T lymphocyte of thymus in the C57BL/6J mouse.
Methods: 50 male Balb/c mice, weighing (18~22)g, were sham or whole-body exposed to EMP at 200kV/m for 400 pulses per day,
which lasted for one week. The thymus from the mice on the 1st d (day) , 3rd d, 7th d, 14th d and 28th d after exposion wad used. The thymus
from two mice of the exposive group and two mice of the sham group were cut and stained with hematoxylin-eosin, then the pathological
changes was detected by optical microscope; The weight of the body and thymus of 3 mice of the exposive group and 3 mice of the sham
group were weighted for the thymus index; The lymphocyte was extracted from the thymus; Then the blood serum was collect to check
out the content of the IL-4 in the peripheral blood. Results: There was no pathological changes on the first and third days after exposion,
however, there was hemorrhage, the structure of thymus changes from the 7th day; The thymus index showed the trend of decline, but
there was no difference between the sham group and the exposive group; The number of T lymphocyte between the sham group and the
exposive group on the 1st,14th and 28th day had no significant deviation, but the difference on the 3rd and 7th day had satistical meaning
(P<0.05); The difference of the content of IL-4 in the peripheral blood between the sham group and the exposive group had no satistical
meaning. Conclusions: The structure of the thymus was destroyed, the thymus index was in the declining trend, the number of T
lymphocyte in the thymus declined, and the content of IL-4 had no difference. EMP had some effects on T lymphocyte in the thymus, but
it is not the sensitive target of EMP on the C57BL/6J mouse. |
