      目的:从c-met 对胰岛β 细胞增殖,细胞周期、糖耐受和对GLUT2 的表达影响三个方面探讨c-met 在胰岛β 细胞功能的 影响及相关机制。方法:在大鼠胰岛β 细胞系INS-1 中运用RNA 干扰技术(RNAi)抑制HGF 的特异性受体c-met 蛋白的表达, 检测其在正常的生理状况下对成熟的胰岛β 细胞增殖以及功能维持的作用。结果:c-met 蛋白对成熟的胰岛β细胞的增殖与周 期并没有显著影响,但对于β 细胞的功能维持具有重要意义。结论:通过调节GLUT2 蛋白来维持β细胞的胰岛素分泌功能,有 助于进一步阐明HGF/c-met 通路在胰岛β 细胞功能损伤的分子机制,从而为糖尿病的预防和治疗提供新的理论依据。
英文摘要:
      Objective: To investigate the effect and mechanism of c-met on the function of pancreatic beta cells in three aspects: β-cell proliferation, cell cycle, glucose tolerance and the impact of GLUT2. Methods: Use RNA interference (RNAi) to inhibit the expression of c-met protein, HGF-specific receptor, in rat pancreatic β cell line INS-1, and detect the effect on the mature β-cell proliferation and function maintenance in normal physiological conditions. Results: There is no significant impact of c-met protein on mature pancreatic β-cell proliferation and cell cycle, but it shows great effect on maintaining the function of β-cell. Conclusion: By regulating GLUT2 protein to maintain β-cell insulin secretion function,contributing to further clarify HGF/c-met pathway in the molecular mechanism of pancreatic β-cell dysfunction, and providing a new theoretical basis on the prevention and treatment of diabetes.
