(n=8),药物干预前1 d 大鼠强迫游泳15 min,药物干预当天,分别腹腔注射相同容积的生理盐水(S 组)、5 mg/kg 氯胺酮(K5 组)、
10 mg/kg 氯胺酮(K10 组)、15 mg/kg 氯胺酮(K15 组)。30 min 后记录大鼠运动能力及不动时间。实验二:20 只Wistar 大鼠随机分
为两组(n=10),所有大鼠均经历21 天慢性不可预知应激试验。第22 天大鼠分别腹腔注射相同容积生理盐水及10 mg/kg 氯胺酮,
于干预前、干预后1 h、2 h、6 h、1 d、4 d、7 d 分别进行敞箱试验,并记录大鼠水平运动及垂直运动得分。结果:与S 组相比,K5、
K10 及K15 组大鼠运动能力无明显变化(P>0.05)且强迫游泳不动时间均显著减少(P<0.01);与干预前生理盐水组相比,生理盐水
干预后1 h、2 h、6 h、1 d、4 d 及7 d 组大鼠敞箱试验水平运动及垂直运动均无明显差异(P>0.05);与干预前氯胺酮组相比,生理盐
水干预后1 h、2 h、6 h、1 d、4 d 及7 d 组大鼠敞箱试验水平运动及垂直运动有明显差异(P<0.05)。结论:在抑郁症大鼠模型中氯胺

Objective: To investigate the antidepressant effects of single administration of ketamine on the depressed rat model.
Methods: Experiment one: Thirty-two rats were randomly divided into four groups (n=8), One day before administration of drugs, rats
were insulted in the forced swimming test (FST) for 15 min. On the second day, rats were injected with the same volume of saline (Group
S), 5 mg/kg ketamine (Group K5), 10 mg/kg ketamine (Group K10), 15 mg/kg (Group K15), respectively. Thirty minutes later, the
locomotor activity and immobility time of rats were recorded. Experiment two: Twenty Wistar rats were randomly divided into two
groups (n=10), all rats were insulted in the chronic unpredictable stress test for 21 d. On the day 22, rats were injected with the same
volume of saline or ketamine 10 mg/kg, respectively. Rats horizontal movement and vertical movement were recorded at the
pre-injection, and the post-injection of 1 h, 2 h, 6 h, 1 d, 4 d, 7 d, respectively. Results: Compared with that in Group S, the locomotor
activity in Group K5, K10 and K15 did not change (P>0.05) but the immobility time of rats (P<0.01). Compared with that in the Group
saline pre-injection, the scores of horizontal movement and vertical movement (P>0.05) in group saline post-injection with 1 h, 2 h, 6 h, 1
d, 4 d and 7 d showed no significant changes; Compared with ketamine pre-injection, Group ketamine post-injection with 1 h, 2 h, 6 h, 1
d, 4 d and 7 d showed significant changes in horizontal movement and vertical movement (P<0.05). Conclusion: Ketamine has exert
fast-acting and enduring antidepressant effect on depressed rat model. |
