      目的:研究TLR4 对脂多糖(LPS)及Polymymin B (PMB)作用下的人骨骼肌细胞的炎症因子表达的影响及其在细胞胰岛素 抵抗中的作用。方法:通过脂多糖(LPS)及Polymymin B (PMB)干预骨骼肌细胞24h,再用胰岛素刺激1h 后,Real-time PCR 检测检 测骨骼肌细胞TLR4、MyD88、TNF-α mRNA 的表达;Western blot 检测TLR4,Myd88 和CRP 的表达;葡萄糖氧化酶法(GOD-POD 法)检测细胞培养液中葡萄糖浓度。结果:TLR4 高表达可以使炎症因子的表达增高,细胞培养液中的葡萄糖浓度增高;TLR4 低表 达可使炎症因子的表达降低,细胞培养液中的葡萄糖浓度没有明显变化。结论:TLR4 调控了炎症因子的表达,继而可以引起胰岛 素敏感性的改变,影响了胰岛素抵抗的发生。
英文摘要:
      Objective: To investigate the effects of TLR4 on the expression of inflammatory factors and insulin resistance in human skeletal muscle cells. Methods: Cultured human skeletal muscle cells were treated by LPS and PMB for 24h, then the cells were incubated in insulin for 1h. The expression of TLR4, Myd88, TNF-α was examined by Real-time PCR and the expression of TLR4, Myd88, CRP were detected byWestern Blot; GOD-POD method was used to assess the concentration of glucose. Results: The activation of TLR4 increased the expression of inflammatory factors and the concentration of glucose; The blocking of TLR4 inhibit the expression of inflammatory factors, the concentration of glucose had no significantly changes. Conclusion: TLR4 regulates the expression of inflammatory factors, then could alter the sensibility of insulin.
