      目的:探究Aβ 低分子量寡聚体对大鼠学习记忆功能的影响;方法:双侧海马内一次性注射Aβ 低分子量寡聚体,15 天后开 始进行行为测试,测试方法采用跳台实验法、穿梭实验法和Morris 水迷宫法;结果:跳台实验结果显示,与对照组比较,Aβ 组跳台 潜伏期短,错误次数多,差异有统计学意义(P<0.05);穿梭实验结果显示,与对照组比较,Aβ 组由明箱进入暗箱的潜伏期短,错误 次数多,差异有统计学意义(P<0.05);水迷宫实验结果显示,与对照组比较,Aβ 组潜伏期长,跨越原平台次数少,差异有统计学意 义(P<0.01);结论:Aβ 组大鼠学习记忆能力和空间分辨能力降低,Aβ 低分子量寡聚体在大鼠体内表现出较强的毒性作用。

      Objective: To investigate the effect of low molecular weight oligomers of β-amyloid peptide on the rat learning and memory. Methods: LMW Aβ oligomers was microinjected into bilateral hippocampus at once. And behavioral tests were carried out after 15 days. Step-down test, Shuttle-box test and Morris water maze test were used to evaluate rat learning and memory function. Results: Step-down test showed that the rats in Aβ made notable errors compared with the control group (P<0.05)and the incubation period (jumping from the platform) significantly reduced(P<0.05). Shuttle-box test showed that compared with the control group, the rats in Aβ made notable errors (P<0.05)and the incubation period (entering in dark chamber from light chamber) significantly reduced(P<0.05). Morris water maze test indicated that the incubation period of finding platform significantly extended (P<0.01)and frequency passing through the prior platform was less in Aβ group (P<0.01). Conclusions: Microinjection of LMW Aβ oligomers into rat hip- pocampus may induce the learning and memory dysfunction, and LWM Aβ oligomers induced an impairment in rat spatial resolution. In a word, LWM Aβ oligomers had strong toxic effects on the rat learning and memory.
