      目的:研究大鼠高动力性肺动脉高压模型转化生长因子-β1( transforming growth factor- beta 1, TGF-β1)和结缔组织生长因 子(connective tissue growing factor, CTGF)的表达变化及意义。方法:45 只SD 大鼠随机分成左肺全切组(A)、假手术组(B)和对照组 (C),每组15 只。手术后6 周测各组大鼠肺动脉平均压(mPAP),右心室肥厚指数(RVH),光镜下检测肺肌型小动脉占肺小血管百 分比(SMA%)。免疫组化观察TGF-β1 和CTGF 在肺组织中的表达,RT- PCR 检测肺组织TGF-β1mRNA 和CTGF mRNA 的表达 水平。结果:①左肺全切后复制了高动力性肺动脉高压模型,引起mPAP、RVH 和SMA%明显增高(P< 0.05)。②左肺全切组 TGF-β1 和CTGF 蛋白及mRNA 表达均较假手术组和对照组显著性增加(P<0.05)。而假手术组和对照组之间各指标没有显著性 差异。结论:TGF-β1 和CTGF 的过度表达是高动力性肺动脉高压发生发展的重要因素,可能共同促进了肺血管重构。

      Objective: To study the expression of transforming growth factor- beta 1 (TGF-β1) and connective tissue growth factor (CTGF) in lung tissues of rats with hemodynamic pulmonary hypertension. Methods: Forty-five SD rats were randomly divided into group A(total resection of left lung), group B(sham operated)and group C (control), with 15 rats in each group. Six weeks later, the mean pulmonary artery pressure (mPAP) and index of right ventricle hypertrophy (RVH) of every group were measured. The percentage of small pulmonary musclarized arteries to all the small pulmonary vessels (SMA%) were observed and calculated under optical microscope. Immunohistochemistry methods were performed to determine TGF -β1 and CTGF protein expression in lung tissue. The expressions of TGF -β1 mRNA and CTGF mRNA in lung tissue were evaluated by RT-PCR. Results: ① The animal model of hemodynamic pulmonary hypertension was confirmed by increased mPAP, index of RVH and SMA% in total resection of left lung group. ② Compared with sham operated group and control group, the expression of protein and mRNA of TGF-β1 and CTGF were significantly increased in total resection of left lung group (P<0.05). But there was no difference in each index between the sham operated group and control group. Conclusion: The TGF-β1 and CTGF are important factors in the development of hemodynamic pulmonary hypertension, and may co-play an important role in the pulmonary artery remodeling.
