      目的:阐明NDRG2(N-Myc downstream-regulated gene 2)在乳腺癌细胞中对CD24 的调控及其对乳腺癌细胞粘附能力的影 响。方法:RT-PCR 和Western blot 方法检测乳腺癌细胞MCF-7 及Bcap-37 中NDRG2 和CD24 的表达;通过腺病毒上调MCF-7 细胞中NDRG2 的表达,或利用siRNA 下调Bcap-37 细胞中NDRG2 的表达,检测CD24 基因和蛋白的变化。粘附实验检测改变 NDRG2 表达水平后对MCF-7 及Bcap-37 细胞粘附能力的影响。结果:MCF-7 细胞中NDRG2 基因和蛋白的表达水平低于 Bcap-37 细胞,而CD24 的表达水平高于Bcap-37 细胞;在MCF-7 细胞中通过腺病毒载体上调NDRG2 可以抑制CD24 的表达并 抑制其粘附能力,而在Bcap-37 细胞中利用siRNA 下调NDRG2 的表达可以提高CD24 的水平及细胞的粘附能力;结论:NDRG2 通过影响CD24 参与调控乳腺癌细胞的粘附能力。
英文摘要:
      Objective: To investigate the effect of N-Myc downstream-regulated gene 2 (NDRG2) on the adhesion potential of breast cancer cells. Methods: Gene and protein expressions of NDRG2 and CD24 were analyzed in MCF-7 and Bcap-37 cells. The expression of CD24 and cell adhesion ability was detected after NDRG2 by adenovirus or siRNA treatment. Results: Both the gene and protein expressions of NDRG2 in MCF-7 cells were lower than Bcap-37 cells. In contrast, CD24 expression is higher in MCF-7 cells compared to that in Bcap-37 cells. Up-regulated NDRG2 expression leads to the decrease in CD24 expression and cell adhesion ability in MCF-7 cells. In contrast, NDRG2 down-regulation results in enhancing the expression of CD24 and cell adhesion potential in Bcap-37 cells. Conclusion: NDRG2, associated with altered CD24 expression, regulates breast cancer cells adhesion ability.
