      目的:探讨stathmin/oncoprotein 18 (Op18)基因在人骨肉瘤中的表达规律及意义。方法:分别采用免疫组化法和Western blot 法检测10 例正常骨组织和56 例骨肉瘤中stathmin 蛋白的表达。结果:免疫组化法检测stathmin 蛋白在正常骨组织、低级别骨肉 瘤(G1 级组)及高级别骨肉瘤(G2 级组)中阳性表达率分别为20 %、65 %、100 %。正常骨组织分别与G1 级组、G2 级组比较,均有显 著性差异(P﹤0.05);G1 级组与G2 级组比较,有显著差异(P<0.05)。Western blot 法检测显示,stathmin 蛋白在正常骨组织、G1 级 组、G2 级组表达相对值分别为(0.34±0.15)、(0.68±0.21)、(0.90±0.17)。正常骨组织分别与G1 级级组、G2 级组比较,差异均有 显著性( P <0.01),G1 级组与G2 级组比较,差异有显著性(P<0.01)。结论:Stathmin 在骨肉瘤中过表达,与骨肉瘤的发生及发展密 切相关。

      Objective: To investigate the expression of stathmin/ oncoprotein 18 (Op18) in human osteosarcoma and its relation to biological behaviors of osteosarcoma. Methods: The stathmin protein expressions of 56 cases with human osteosarcoma and 10 cases of normal bone tissues were examined by immunohistochemical staining and Western blot methods. Results: The positive rate of stathmin in normal human bone tissues, G1 grade and G2 grade were 20%, 65%, 100% respectively. The difference between normal human bone tissues and G1 grade, G2 grade were significant (P<0.05)respectively; the difference between G1 grade and G2 grade were significant(P<0. 05). Expression of stathmin in normal human bone tissues,G1 grade and G2 grade were (0.34±0.15),(0.68±0.21),(0.90±0.17) respectively. The difference be- tween normal human bone tissues and G1 grade, G2 grade were significant (P<0.01) respectively; the difference between G1 grade and G2 grade were significant (P<0.01). Conclusion: Overexpression of stathmin plays an important role in development and progression of human osteosarcoma.
