      目的:探讨Bmi1 在肝细胞肝癌(HCC)中的表达及与增殖和凋亡的关系。方法:收集HCC 标本54 例及相应的癌旁组织,10 例正常肝组织标本,采用免疫组织化学EnVision 二步法显示Bmi1 的表达并结合增殖与凋亡特征进行分析。结果:在肝癌组织、癌 旁组织细胞中Bmi1 表达定位于胞核中,阳性表达率分别为79.6%(43/54),31.2% (17/54),10 例正常肝组织未见表达,3 组差异有 统计学意义(P<0.005)。HCC 中Bmi1 的高表达与年龄、性别、肿瘤大小、肿瘤数目、临床TNM 分期、是否有肝硬化及是否有 HBsAg 感染无显著相关(P>0.05),但与组织学分级有关,高、中分化组Bmi1 表达率显著高于低分化组( P<0.05)。肝癌Bmi1 阳性 组增殖指数(PI)(50.3±21.4)%显著高于阴性表达组(17.3±7.1)%(P<0.05),凋亡指数(AI)无明显差异(P>0.05)。结论:Bmi1 在HCC 中高表达,其表达增高可能与HCC 的进展相关。

      Objective: To investigate the expression of Bmi1 and its relationship with cell proliferation and apoptosis in hepatocellular carcinoma (HCC). Methods:54 cases of HCC tissues and paraneoplastic hepatic tissues and 10 normal liver tissues were selected for the study. These samples were detected by EnVision immunohistochemical and TUNEL methods. All data were processed by SPSS 12.0 version in computer. Results:The expression of Bmi1 was localized in nucleus of HCC tissues and paraneoplastic liver tissues, showing yellowish-brown. Bmi1 was expressed in 43 of the 54 cases of HCC (79.6%); 17 of the 54 cases of paraneoplastic tissues (31.2%) and negative in the all of normal liver tissues. The rate of Bmi1 positive expression was significantly higher in HCC than that in paraneoplastic tissues and normal liver tissues (P<0.005). The increased Bmi1 expression in HCC was not correlated with gender, age, tumor size, tumor number, portal involvement, TNM stage, liver cirrhosis and HBsAg status (P>0.05). However, the expression of Bmi1 in HCC was associated with the histological differentiation, of which well and moderately differentiated HCCs were higher than that in poor differentiated HCCs (P>0.05). The average value of proliferative index (PI) in positive cases in HCC was (50.3±21.4) %, and (17.3±7.1)% in negative cases(P<0.05). The average value of apoptosis index(AI) in Bmi1 positive cases in HCC was (10.4±1.4)%, and (9.6±2.2)% in negative cases (P>0.05). Conclusion:There is an overexpression of Bmi1 in HCC, which may be related with the development and progression of HCC and may be involved in the regulation of cell proliferation of HCC.
