      目的:探讨胃癌患者血管内皮生长因子C(vascular endothelial growth factor-C, VEGF-C 及血管内皮生长因子受体-3(vascular endothelial growth factor receptor-3,VEGFR-3)在胃癌组织中的表达,从而确定胃癌预后的分子标志物。方法:搜集整理临床资料, 采用Real-time PCR 及ELISA 法检测43 例胃癌组织VEGF-C 和VEGFR-3 的表达。结果:43 例胃癌组织中均有不同程度的 VEGF-C 和VEGFR-3 的表达,Real-time PCR 结果显示胃癌组织淋巴结转移组和非转移组VEGF-C 和VEGFR-3 的表达分别为 0.07±0.01 和0.12±0.01,0.03±0.01 和0.06±0.02,与正常对照组相比,差异有显著性(p<0.05)。ELISA 检测显示,与正常胃组织 中VEGF-C 和VEGFR-3 的蛋白表达相比,胃癌无淋巴结转移组及胃癌并发淋巴结转移组中VEGF-C 和VEGFR-3 均明显增加。 结论:VEGF-C 和VEGFR-3 的表达与胃癌淋巴结转移密切相关,提示胃癌标本VEGF-C 和VEGFR-3 的检测可作为胃癌预后的 分子标志物。

      Objective: To investigate the expression of vascular endothelial growth factor C (VEGF-C) and Vascular endothelial growth factor receptor -3 (VEGFR-3) in gastric cancer, so that to determine the molecular markers in prognosis of gastric cancer. Methods: Collecting clinical data methods, Real-time PCR and ELISA were used to detect the expression of VEGF-C and VEGFR-3 in 43 cases of gastric cancer. Results: 43 cases of gastric cancer have different levels of VEGF-C and VEGFR-3. Real-time PCR results showed the VEGF-C and VEGFR-3 expression were 0.07±0.01 and 0.12±0.01, 0.03±0.01 and 0.06±0.02 respectively in lymph node metastasis group and non- metastasis group in gastric cancer., the difference was statistically significant when compared with the control group (p <0.05). ELISA examination demonstrated that expressions of VEGF-C and VEGFR-3 protein in the gastric cancer group without lymph node metastasis and the gastric cancer group complicated by lymph node metastasis were obviously increased when compared with that in normal gastric tissue. Conclusions: VEGF-C and VEGFR-3 expression were closely related with lymph node metastasis, indicating that VEGF-C and VEGFR-3 detection can be used as molecular markers in prognosis of gastric cancer.
