进细胞增殖活性,而高浓度阿司匹林能够强烈抑制细胞增殖。0.5、1、2 mmol/L浓度阿司匹林可促进BMSCs 的成骨性分化,中低

Objective: To investigate the effect of aspirin on differentiation of marrow stromal cells (MSCs) into osteoblast in vitro.
Methods: Bone marrow stromal cells of 7 days old SD rats were cultured. After 3 times of passages, the cells were induced to osteogenic
differentiate in mediums that contained 0.5, 1, 2, 5,10mmol/L aspirin separately, and control group was establish. The cell proliferation
was assayed by CCK-8 method. The osteogenic differentiation markers including alkaline phosphatase (ALP) activity, osteocalcin secretion
and mineralized bone modulus between the Aspirin groups and the control were compared. Results: Aspirin didn't stimulate the proliferation
of BMSCs, and high concentration of aspirin had the inhibited effect. 0.5, 1, 2 mmol/L aspirin stimulated the osteogenic differentiation
of BMSCs. In different phase, alkaline phosphatase activity and osteocalcin secretory volume in low and medium concentration
of aspirin groups was higher than that of the control group. Alizarin red stain for calcium nodus in aspirin groups was far more than that
in control group in 14 days. Conclusions: Low and medium concentration of aspirin stimulated the osteogenic differentiation of BMSCs,
which indicated that aspirin had the ability to enhance the bone metabolism activity. |
