摘要 目的:探究乳腺癌组织中微小RNA-17-3p(miR-17-3p)、微小RNA-671-3p(miR-671-3p)表达变化与临床病理特征、细胞增殖基因YAP1、Piwil2与细胞侵袭基因Rab11、TUG1表达的关系。方法:选取河南科技大学第一附属医院在2022年1月-2023年1月期间收治的160例乳腺癌患者,取手术切除的癌组织和癌旁正常组织。对比乳腺癌组织、癌旁正常组织以及不同临床病理特征的miR-17-3p mRNA、miR-671-3p mRNA相对表达量,对比乳腺癌组织、癌旁正常组织的细胞增殖基因YAP1、Piwil2与细胞侵袭基因Rab11、TUG1相对表达量。采用Pearson相关系数分析乳腺癌组织miR-17-3p mRNA、miR-671-3p mRNA与细胞增殖基因YAP1、Piwil2,细胞侵袭基因Rab11、TUG1的相关性。结果:对比癌旁正常组织,乳腺癌组织的miR-17-3p mRNA相对表达量明显升高且miR-671-3p mRNA相对表达量明显降低(P<0.05)。对比TNMⅠ期、无腋窝淋巴结转移、Ki-67阴性,TNMⅡ期、有腋窝淋巴结转移、Ki-67阳性乳腺癌组织的miR-17-3p mRNA明显更高且miR-671-3p mRNA表达水平明显更低(P<0.05)。乳腺癌组织中细胞增殖基因YAP1 mRNA、Piwil2 mRNA,细胞侵袭基因Rab11 mRNA、TUG1 mRNA表达量比癌旁正常组织高(P<0.05)。miR-17-3p mRNA表达与YAP1 mRNA、Piwil2 mRNA、Rab11 mRNA、TUG1 mRNA表达均呈正相关(P<0.05),而miR-671-3p mRNA表达与YAP1 mRNA、Piwil2 mRNA、Rab11 mRNA、TUG1 mRNA表达均呈负相关(P<0.05)。结论:miR-17-3p、miR-671-3p在乳腺癌中高表达,且与腋窝淋巴结转移、TNM分期、Ki-67阳性相关。在乳腺癌细胞增殖、细胞侵袭过程中miR-17-3p、miR-671-3p均有参与并发挥重要作用。 |

ABSTRACT Objective: To explore the relationship between the expression changes of microRNA-17-3p (miR-17-3p) and microRNA-671-3p (miR-671-3p) and clinicopathological characteristics, the expression of cell proliferation genes YAP1 and Piwil2, and cell invasion genes Rab11 and TUG1 in breast cancer tissues. Methods: 160 patients with breast cancer who were admitted to The First Affiliated Hospital of Henan University of Science and Technology from January 2022 to January 2023 were selected, and the cancer tissue and adjacent normal tissue resected by surgery was taken. The relative expression levels of miR-17-3p mRNA and miR-671-3p mRNA in breast cancer tissue, adjacent normal tissue and different clinicopathologic characteristics were compared, and the relative expression levels of cell proliferation genes YAP1 and Piwil2 and cell invasion genes Rab11 and TUG1 in breast cancer tissue and adjacent normal tissue were compared. Pearson correlation coefficient was used to analyze the correlation between miR-17-3p mRNA, miR-671-3p mRNA, and cell proliferation genes YAP1 and Piwil2, as well as cell invasion genes Rab11 and TUG1 in breast cancer tissue. Results: Compared with the adjacent normal tissue, the relative expression level of miR-17-3p mRNA was significantly increased and the relative expression level of miR-671-3p mRNA was significantly decreased in the breast cancer tissue (P<0.05). Compared with TNM stage I, no axillary lymph node metastasis, Ki-67 negative, TNM stage II, axillary lymph node metastasis, Ki-67 positive of breast cancer tissue significantly higher miR-17-3p mRNA and significantly lower miR-671-3p mRNA expression level (P<0.05). The expression levels of cell proliferation genes YAP1 mRNA, Piwil2 mRNA, cell invasion genes Rab11 mRNA and TUG1 mRNA in the breast cancer tissue were significantly higher than those in the adjacent normal tissue (P<0.05). miR-17-3p mRNA expression was positively correlated with YAP1 mRNA, Piwil2 mRNA, Rab11 mRNA and TUG1 mRNA expression (P<0.05). miR-671-3p mRNA expression was negatively correlated with YAP1 mRNA, Piwil2 mRNA, Rab11 mRNA and TUG1 mRNA expression(P<0.05). Conclusion: miR-17-3p and miR-671-3p are high expressed in breast cancer, and are correlated with axillary lymph node metastasis, TNM staging, and Ki-67 positive. Both miR-17-3p and miR-671-3p are involved and play an important role in the process of breast cancer cell proliferation and cell invasion. |
